| Objective: To summarize the medication rules and academic ideas of Professor Deng Yunming in the treatment of osteoporosis by using the TCM inheritance computing platform V3.0 and network pharmacology,and to provide clinical basis for the internal administration of Chinese medicine for the treatment of this disease.method:1.TCMSP inheritance auxiliary platform: The research subjects were selected as osteoporosis patients in the outpatient department of Professor Deng Yunming of Jiangxi University of Traditional Chinese Medicine from May 1,2019 to May 1,2022,Record the medical record.2.Network pharmacology: Conduct network pharmacology research on core prescriptions excavated from the previous step.The main steps are:(1)to find the core prescription active ingredients and corresponding targets of OB≥30% and DL≥0.18 in the TCMSP and Swiss Target Prediction databases.(2)Gene Cards and OMIM were used to search for disease targets,and the intersection genes of core prescriptions and disease common targets were constructed.(3)Use Cytoscape 3.9.1software to construct a "traditional Chinese medicine-ingredient-target network diagram".(4)The STRING platform was used to construct the PPI network,and then imported into Cytoscape for topological analysis to find the key targets of core prescription therapy OP.(5)Use DAVAD software to export GO and KEGG pathway enrichment analysis,and use the microbiotic database to draw histograms and bubble chartsResults: 1.TCM inheritance support platform:609 data of Professor Deng Yunming who met the inclusion criteria for the identification and treatment of osteoporosis were analyzed.Common symptoms are: fixed sores,joint deformity,joint tingling,unfavorable movement,dull complexion,dull joint pain,sore waist and knees,unfavorable pitching and turning,dizziness,and sluggishness.Common types are: blood stasis block,renal deficiency,kidney qi deficiency,spleen deficiency.Common pathogenesis: blood stasis block as the target,kidney deficiency as the basis,disease is the virtual standard,and the virtual and real are sandwiched.Common treatments: relieving pain,activating blood circulation and removing stasis,warming the kidney,invigorating the essence and activating the blood,tonifying the kidney and qi,strengthening the spleen and stomach,etc.;Commonly used drugs: astragalus,angelica,Xu Changqing,ox knee,danshen,Wujia peel,eucommia,licorice,goji berry,chicken blood vine,calcined keel,calcined oyster,bone crushed tonic,dodder seed,deer antler cream,suoyang,poria,baiju,etc.Three core combinations were extracted by K-means algorithm and regression model cluster analysis: combination 1—licorice,astragalus,angelica,Xu Changqing,and white peony;Combination 2-angelica,deer antler cream,five-plus peel,dodder seed,calcined keel;Combination 3-licorice,astragalus,angelica,poria,white peony;2.Network pharmacology:(1)through the search of TCMSP and Swiss Target Prediction platform,106 active ingredients of core prescription 1 and 241 targets;41active ingredients of core prescription 2,323 targets,108 active ingredients of core prescription 3 and 243 targets.(2)A total of 4688 OP-related disease targets were obtained in Gene Cards and OMIM databases.After intersection mapping,147 core prescription 1 and disease intersection targets,187 core prescription 2 and disease intersection targets,and 148 core prescription 3 and disease intersection targets were obtained.(3)Introduce Cytoscape to build the core prescription TCM-component-target network.PPI network map was constructed and network topology analysis was conducted to screen the key targets of core prescription for OP:AKT 1,STAT 3,CXCL 8,TP 53,VE GF A,MMP-9,etc.(4)Performed the GO and KEGG functional enrichment analysis,In terms of the molecular function,Molecular targets mainly affect the enzyme binding,protein kinase binding and protein homodimerization activity;In terms of the cellular composition,Molecular targets to form the receptor complex,plasma membrane microcapsule and cell solutes;In terms of the biological processes,These targets mainly involve response to drugs,senescence and positive regulation of apoptotic processes,KEGG mainly involves the involvement of signaling pathways such as AGE-RAGE,IL-17,PI3K-Akt,TNF,and MAPK.3.Conclusions: 1.Traditional Chinese medicine inheritance computing platform:(1)Using the TCM inheritance support platform,the cases and prescriptions of famous and old TCM practitioners can be analyzed more systematically and objectively from different levels and aspects,which has important guiding significance for clinical work.(2)After systematic statistical screening,it was concluded that Deng Lao’s treatment of osteoporosis was divided into 3 types: stasis block,kidney deficiency,and spleen and kidney yang deficiency(3)Deng Lao believed that osteoporosis kidney deficiency was based on myelin deficiency,blood stasis block was the standard,and astragalus,angelica,Xu Changqing,ox knee,danshen,Wujiapi,and eucommia were the medicines reused by his prescription,and on this basis,the clinical evidence was added and subtracted;The treatment is mainly to warm the kidney,activate blood and remove stasis,and strengthen the spleen and stomach.2.Network pharmacology: Core prescription regulates AKT 1,STAT 3,CXCL 8,TP 53,VE GF A,quercetin,kaempferol,β-sitosterol,and catechin in AGE-RAGE,TNF,IL-17,MAPK and other signaling pathways,which mainly involve apoptosis,autophagy,inflammatory response,cellular aging,immune regulation and other biological processes,so as to play a role in treating OP.It embodies the characteristics of multi-component,multi-target and multi-pathway of TCM compound network pharmacology. |
