Objective: Angiogenesis is the propelling force for tumor growth and metadtasis, and antiangiogenic therapy represents one of the most promising modalities for cancer treatment. At present, pan marker of vascular endothelial cells(EC) such as CD34, CD31 and vWF are used in the research of tumor angiogenesis. Due to the high heterogeneity of the ECs, and the affinity of the above antibodies to the commen ECs, these antibodies are lack of specificity to the neovascular ECs in tumor tissues. In this research, CD105 acted as a new marker of ECs to identify the new born vessels in the tumor tissues in order to contribute a new method to the antiangiogenic research and therapy. We also identified the expression of Nestin in liver cancers to explore its feasibility as a new marker of neovascularization. Method: Double immunohistochemistry was used to identify the expression of CD105, Nestin and α-SM-actin in the fresh tumor and peri-tumor tissues of 34 cases of liver cancer, and 2 cases of hemangioma , and 8 cases of normal liver tissue; Immunofluorescence was applied to identify the difference of expression between CD105 and Nestin. Result: There are significant differences in the expression of CD105 between the tumor tissues and non-tumor tissues of the same patient (P<0. 001), and so with the expression of Nestin(P<0. 001) ; Statistic significance are detected between the expression of CD105 and Nestin in the tumor tissues of the same patient, with higher expression of the former; the expression of CD105 is closely related to portal vein embolus, capsule invasion andmetastasis within the liver(p1=0.009; p2<0.001; p3=0.001)but not such a case with Nestin.Conclusion: 1 When malignant solid tumors grow to certain stage the intensity of microvessles in tumor tissues will seemingly exceed that of the according benige tissues. 2 CD105 is abundantly expressed in angiogenic endothelial cells(ECs) of both peritumoral and intratumoral vessels. 3 The results of our study indicate that CD105, as a marker of angiogenic ECs, may be applied to the research of antiangiogenesis and bear theraputic potential in patients with liver cancer; Although Nestin is also highly expressed in angiogenic ECs it has little relation to capsule invasion and metastasis within the liver. So its clinical application needs to be further investigated. 4 It' s much more seemingly to detect new born vessels in tumor tissues with double immunohistochemistic staining for a -SM-actin and markers of angiogenesis.
