| P27 is a anticancer gene which was discovered by Polyak in 1994 and it is a hot stability protein. P27 control cell cycle by two ways , one is to restrain the activation of CDK , the other is to restrain the activity of cyclin-CDK of post- activation and then the course of cell cycle was controled . p27 participate in cell differentiation and witherness besides participating in the cell cycle . the restraint activity of p27 is ascendent when p27 is location in the cell nucleus and the expression of p27 in cell plasm will reduce the period of cell suvival . the low expression of p27 in many carcinomas is distinctly correlated with prognosis and high expression of p27 can distinctly restrain growth of the carcinoma , so that artifical p27 as a aim gene is capbable of treating carcinoma . Bcl-2 gene locate in chromosome 18q2 , the excess expression of it was discovered in follicle lymphoma firstly . bcl-2 control arisement or development of carcinoma via preventing apoptosis of cell . excess expression of bcl-2 can prevent cell apoptosis, consequently lead to the imbalance between the proliferation and apoptosis. Though speed of cell proliferation doesn't change , reducment of apoptosis cell result in relative increasement of cell number so that prolong the life of cell . the relationship between expression of bcl-2 and prognosis of carcinoma has different conclusion in different type of carcinoma . some research indicate that lack of bcl-2 has no distinct effect to cell funtion , body has some endurence to lack of bcl-2 . Some literature reported that p27 has distinct correlation with prognosis of carcinoma . but if p27 can diagnose early stage cancer there have different standpoint , some literature consider bcl-2 can diagnose early carcinoma , but if bcl-2 corelated with prognosis there have different standpoint , so this text combine the two factor and inspect them toghter to see the effect of the two factor in diagnosing early endometrial cacinoma ,comfirming degree of malignency and forcasting prognosis .Objective: to study the expression of p27 and bcl-2 in the endometrial hyperplasia , endometrial carcinoma and the relationship between the expression and the clinic pathological parameter , than examine the significance of the occurrence, development,prognosis of the carcinoma. Methods: S-P method was used to examine the expression of the p27 and bcl-2 in the 24 endometrial (simple and complex) hyperplasia , 20 endometrial atypical hyperplasia and 56 endometrial carcinoma . Results: 1. the expression of p27 is located in the cell nucleus and cell plasma , the expression of p27 is 75%,65%,53.6% respectively in the three sets , and descended gradually , there was no significance difference among the three sets (p>0.05) . the expression of bcl-2 is also located in the cell nucleus and cell plasma ,the expression of the bcl-2 is 79.2%,45.0%,44.6% respectively in the three sets , to match with atypical hyperplasia and endometrial carcinoma, the expression of p27 in the simple hyperplasia and complex hyperplasia has a significant difference (p<0.05) , there was no significance difference between atypical hyperplasia and endometrial carcinoma (p>0.05). 2. the expression of p27 is related with the clinical stage ,histological grade and muscle invasion (p<0.05), and the expression is not related with pathological type and lymphnode metastasis (p>0.05) , the expression of bcl-2 is related to lymphnode metastasis (p<0.05) and not related with other clinic pathological parameter(p>0.05). 3. the expression of p27 and bcl-2 is positive correlated , correlation coefficient rs=0.332(p<0.05) Conclusion: p27 and bcl-2 play roles in different stage of the development of the endometrial carcinoma . bcl-2 may predicting the occurrence of the endometrial carcinoma in the early stage , the low expression of p27 is associated with the development of endometrial carcinoma and may be regarded as a useful index for prognosis . together inspected p27and bcl-2 has important effect... |
