Objective: To investigate the significance of CD34 and CD105 as the endothelial markers, and to discuss the expression of VEGF, COX-2, HIF-1 α and the relation to clinical pathological character and angiogenesis in colorectal carcinoma. Methods: The expression of CD34, CD105 , VEGF, COX-2 and HIF-1 α was detected by immunohistochemistry in 78 cases of colorectal carcinoma. Results: (1)The relationship between the two groups of MVD counting by CD34 and CD105 staining had significant difference, the MVD counting by CD105 staining was less than that by CD34 staining(t=4.782, P<0.001). (2)Of all cases the expression of VEGF. COX-2. HIF-1 α was 75.6%. 70.5%. 69.2% respectively, the MVD counting in positive groups was higher than that in negative groups(t =2.372, t =2.368, t =2.272, P<0.05). (3)The expression of VEGF had statistical significance among different invasive depth, metastasis and non-metastasis groups ( x ~2=5.374, P<0.05, P =0.032); the expression of COX-2 between the lymph node metastasis and non-lymph node metastasis had remarkble significance, so did in different Dukes stages ( x ~2=5.468, x ~2=5.654, P<0.05); the expression of HIF-1 α also had statistical significance in different invasive depth groups and in different Dukes stages( x ~2=4.397, x ~2=4.255, P<0.05). (4)The expression of HIF-1 α and COX-2 was positive correlation with the expression of VEGF(r =0.463, P<0.001, r =0.288, P<0.05). Conclusion: (1)CD34 was a kind of pan-vascular endothelial cell markers and CD105 only strongly expressed on the endothelial cells of newborn blood vessels, so CD105 was obviously superior to CD34 in detecting the newborn blood vessels of tumors. (2)The expression of VEGF. COX-2. HIF-1 α was positive correlation with MVD in colorectal carcinoma. To inhibit their expression might be an approach to inhibit angiogenesis in colorectal carcinoma. (3)The expression of VEGF correlated with invasive depth and metastasis of colorectal carcinoma; The expression of COX-2 correlated with lymph node metastasis and Dukes stage of colorectal carcinoma; The expression of HIF-1 α correlated with invasive depth and Dukes stage of colorectal carcinoma. Detecting the expression of VEGF. COX-2. HIF-1 α in colorectal carcinoma was valuable to estimate the clinical pathological stage of colorectal carcinoma. (4)The expression of COX-2. HIF-1 α was positive correlation with VEGF.
