Effect Of Renewed Xuefu Zhuyu Decoction On Function Of Endothelium And Myocardial Fibrosis In Renovascular Hypertensive Rats

Objective To study the Effect of Renewed Xuefu Zhuyu Decoction on Function of Endothelium And myocardial fibrosis in Renovascular Hypertensive Rats,and illustrate the relationship between improvement of Function of Endothelium and formation of myocardial fibrosis.Methods Forty rats of ten weeks old were divided into five groups:Model group (hypertensive rats group),Captopril group,Renewed Xuefu Zhuyu Decoction group,sham operated group,control group.There were eight rats in each group and the mean body weigh of each rat is 180±17g.The Renovascular Hypertensive Rats model of abdominal aorta stegnosis was made and then blood pressure was measured through tail-cuff manometer.The drugs was given through intragastric administration in each group.Each rat in Captopril group was given Captopril 100mg/kg and in Renewed Xuefu Zhuyu Decoction group 3ml/d for eight weeks;each rat in other three groups was given distilled water 3ml/d for eight weeks too.vWF,ET,D-D in blood plasm was measured using ELISA(enzyme linked immunosorbent assay) and LVWI(left ventricular mass index) was also measured;total collagen were analyzed by Hematoxylin-Eosin staining and computerized image analysis system.Collagen typeⅠand Collagen typeⅢwere measured by immunohistochemistry.Result Compared with Hypertensive rats group(model group),vWF,ET,D-D in blood plasm of Renewed Xuefu Zhuyu Decoction group was lower(P＜0.01);In Renewed Xuefu Zhuyu Decoction group and Captopril group the blood pressure was also lower and LVWI of Renewed Xuefu Zhuyu Decoction group and Captopril group was decreased (P＜0.01).CollagenⅠwere significantly higher in Model group than in Renewed Xuefu Zhuyu Decoction group and Captopril group(P＜0.01).The collagenⅢwas also significantly increased in Model group while Renewed Xuefu Zhuyu Decoction group and captopril group decreased significantly(P＜0.01). Conclusion Renewed Xuefu Zhuyu Decoction can improve Function of Endothelium and prethrombotic state in Renovascular Hypertensive Rats and prevent the formation and development of myocardial fibrosis;There is a positive linear correlation between the improved Function of Endothelium and the development of myocardial fibrosis.
