Experimental Research On Rabbit Model Of Congestive Heart Failure (CHF) Induced By Adriamycin Treated By Warming Yang And Diuretic

Objective:TO explore the effects of Wenyang Qiangxin decoction (WQD) in different dose on heart function,neural endocrine,cardiac structure,myocyte apoptosis of the experimental rabbit with chronic congestive heart failure.Methods:50 experimental rabbits were numbered and divided into 2 parts randomly:the normal group with 10 rabbits and the left 40 rabbits which will be model with chronic congestive heart failure by injecting Azithromycin twice a week for 8 weeks.In the 9th week, to know the changes of heart functions and cytokine in blood, the rabbits were tested by serological means and evaluated by UCG, then the left 40 rabbits were divided into four groups by the heart functions which were evaluated by. UCG:high dosage group,low dosage group,control group,model group.The four groups were treated with high and low doses of WQD,Captopril,distilled water respectively once a day, the treatment lasted for 4 weeks. The each dose of drug was 10ml/Kg, The results of UCG,Ang-II,ANP,cardiac structure and pathological tests,myocyte apoptosis (Bax,Bcl-2) were measured.Results:(1) Compared with normal group in the 9th week, the heart function was declined significantly by UCG (P<0.01, P<0.05) the levels of some serum index (Ang-â…¡,ANP) were increased by blood testing (P<0.01). (2) After treatment:Compared with model group the heart function of model group was improved significantly, the levels of some serum index (Ang-â…¡,ANP) were decreased (P<0.01, P<0.05); Compared with the model group, There were significant differences in Pathological section results and myocyte apoptosis testing(P<0.01, P<0.05). There was not significant difference in every index between the high doses group and Captopril group(p>0.05), and was better than the control group the low dose group.Conclusion:WQD in both light and strong doses and Captopril group can improve the function of the heart,prevent Ventricular Remodeling,regulate the level of the neural endocrine, inhibit the expressing way of apoptosis, strong dose is better than light dose.
