| Radix Palygalae was first recorded in Shennong Bencaojing with long history in use for medicine,which is the dried roots of Polygala tenuifolia Willd.and Polygala sibirica L. as described in Chinese Pharmacopoeia 2010 Edition. Radix Palygalae is bitter,pungent and warm in nature,it passes through Heart, kidney and lung meridian. It has the effect of tranquilizing the mind,promoting intelligence,coordinating heart-kidney,expelling phlegm and subsiding swelling.It was used to relieve symptoms like insomnia and dreamful sleep caused by heart yang kidney yin incoordinating,forgetful, palpitates,staring spells,expectoration,sores ulceration,acute mastitis.However,some adverse reaction like slightly nauseated and vomiting with routine dosage of Radix Palygalae were found clinically.Modern research has found that saponins which_ can irritate the stomach lining is the toxic ingredients of Radix Palygalae.To explore the cause and mechanics of the adverse reaction of Radix Palygalae, this study analyze the difference content of the saponins in the different processed products of Radix Polygalae(crude Radix Polygalae,licorice And Honey Processed Radix Polygalae), and their acute toxicity,for providing a scientific basis for establishing rational preparing process and safe, effective using in clinical practice.Chapters 1 Literature ReviewIn this chapter,five reviews are reviewed by quoting 107 documents together:the research of Radix Polygalae on the history successive changes, chemical composition, pharmacological effects, the extraction process and The modern research development of preparing.Chapters 2 Develop a method of detecting the active contents of polygalacic acid and senegenin in Radix Polygalae,and do the research methodology.Objiective:To establish a RP-HPLC method for determining the saponins in Radix Polygalae.Methods:The content of polygalacic acid and senegenin were determined by RP-HPLC. Agilent ZORBAX SB-C18(4.6mm×250mm,5μm)at 25℃was used with a mobile phase of acetonitrile-0.1% phosphoric acid(41:59) and detected at 210 nm.The flow rate was 1.Oml-min-1.Results:the linear range of polygalacic acid was 0.420~4.200μg,with a correlation coefficient of 0.9999,the linear range of senegenin was 0.440~4.400μg,with a correlation coefficient of 1,the average recovery of polygalacic acid was 98.06%(n= 6,RSD=1.20),the average recovery of senegenin was 95.02%(n=6,RSD=1.18).Conclusion:The method is simple and accurate,with good repeatability and can be used to control the content the saponins in Radix Polygalae.Chapters 3 Analyze the difference contents of the saponins in the different processed products of Radix Polygalae(crude Radix Polygalae,licorice And Honey Processed Radix Polygalae).Objective:To analyze the difference of the saponins in the different processed products of Radix Polygalae(crude Radix Polygalae,licorice And Honey Processed Radix Polygalae).Methods:To determine the content of Polygalacic acid and senegenin in the different processed products of Radix Polygalae by HPLC. using an Agilent ZORBAX SB-C18(250mmx4.6mm,5μm), with acetonitrile-0.1% phosphoric acid (41:59) as the mobile phase,the flow rate was 1.0 mL-min-1,temperatures was 25℃and UV detection wavelength at 210nm.Results:The order of the content of Polygalacic acid and senegenin (from higher to lower) was licorice Processed Radix Polygalae,crude Radix Polygalae,Honey Processed Radix Polygalae.Conclusion:The different process method have some effects on the contents of saponins in Radix Polygalae. The content of Polygalacic acid and senegenin in Honey Processed Radix Polygalae was the lowest.Chapters 4 The Doehlert design with a desirability,which allowed a sequential response surface methodology to be performed,was used to study the extracting technology of the saponins in Radix Polygalae.Objective:To study the optimum extracting technology of the saponins from Radix Polygalae.Methods:ethanol concentration,ethanol volume and extracting times as independent variables,yields,the extraction rate of Polygalacic acid and senegenin as responses, RSM with Doehlert design was adopted to explore the optimum extracting technology of the saponins from Radix Polygalae.Results:The optimum extracting technology were as follows:refluxing and extacting with 10-fold 60% alcohol for 3 times and 2 h for each time.Conclusion:The Doehlert design with a desirability,which allowed a sequential response surface methodology to be performed,is a methods easy to be used with good predictability.It can straightforwardly and visually describe the function relation between the independent variables and responses,and the results can accurate reflect the reality.Chapters 5 To study the acute toxicity of the different processed products of Radix Polygalae(crude Radix Polygalae,Iicorice And Honey Processed Radix Polygalae).Objective: To study the acute toxicity of the water extracts (ERWB) and 60% ethanol extracts (EREE) from the different processed products of Radix Polygalae(crude Radix Polygalae,licorice And Honey Processed Radix Polygalae).Methods:The ERWE and EREE were prepared from the different processed products of Radix Polygalae and determined the content of saponins respectively. Poisoning condition and death condition of the mices intragastriced with The ERWE and EREE were observed within fourteen days,and modified Karber's method was used to calculate LD50 and 95% confidence.Results:the EREE of licorice Processed Radix Polygalae has the maximum toxicity(LD50=3.120±0.439g-kg-1) with highest content of saponins(22.0894mg-m1-1'), and the ERWE of Honey Processed Radix Polygalae has the minimum toxicity (LD50=14.976±2.11Og-kg-1)with lowest content of saponins(6.1745mg-m1-1).Conclusion:different processing methods have effects on the contents of saponins in Radix Polygalae.The experiments show that the toxicity of Radix Polygalae is in direct proportion to the content of saponins. |
