According to traditional research method of Radix Eleutherococci, this study researched the preparative separation, purification and physical structural modifications of main medical composition in Radix Eleutherococci. As a result, an economical and efficient technical route is founded. Based on this route, related products of Radix Eleutherococci will be creatively developed.1. Three main phenol glycosides and aglycones (syringin, eleutheroside E and isofraxidin) in Radix Eleutherococci are extracted by the process of homogenization and HPLC is used for analyzing and determining their content. The extraction rate and purity of three targeted compositions were as follows:syringin,0.0016% and 0.052%; isofraxidin,0.018% and 0.68%; eleutheroside E,0.035%, and 1.10%, respectively.2. After the process of homogenization extraction, extraction solution contains three targeted compositions, Acanthopanax senticosus polysaccharide and so on. Due to the negative effect of Acanthopanax senticosus polysaccharide in the physical structural modifications process, macroporous resin was used for cleaning the Acanthopanax senticosus polysaccharide from the extraction solution. HPLC is used for analyzing and determining. Through this process, the main medical compositions are purified. In fact, the contents of syringin, eleutheroside E and isofraxidin increased 26-fold,37-fold and 24-fold and their recoveries were 80.93%,93.97% and 93.79%, respectively.3. After the macroporous resin process, the product was modified by spry-drying method. The processed product was characterized by Scanning Electron Microscopy (SEM), Derivative Thermogravimetry (DTG), Thermogravimetry (TG) and X-Ray Diffraction (XRD) analyses. Average particle size of the product containing main meidcal compositions was significantly decreasing, and average particle size of the processed product was the 1-5μm.
