This dissertation reported the investigation on the chemical constituents and their activation for PXR-mediated CYP3A4expression, cytotoxic activities and anti-HIV-1activities of G resinaceum and and G. cochlear.As a member of Gandoderma Karst., G. resinaceum Boud.(Polyporacea) has long been used for enhancement of internal balance and functions of immune system. Researches showed that it possessed antibacterials and antioxidant effects. However, chemical constituents of G. resinaceum were studied rarely. To investigate the chemical constituents and study their biological activities of the fruiting bodies of the fungus,18compounds, including14triterpenoids,3ergostane sterols and1prenylate phenolic compounds were isolated. Among them,8compounds (1-8) were new triterpenoids. Sixnorlanostane triterpneoids and lanostane triterpenoids with the cleavage of double bond between C-8and C-9were identified for the fist time. The activation for PXR-mediated CYP3A4expression of these compounds was evaluated. The results showed that ganoderic acid Y (9), ganoderol B (10) and lucidone B (12) were inducers for PXR-mediated CYP3A4expression. And their activities were stronger than positive control (Rifampin).G. Cochlear (Blume et Nees) Bres. was another Ganodema species. Investigation of chemical constituents of G. cochlear showed that four ergostane sterols, fifteen lanostane triterpenoids, and five prenylate phenolics were isolated. Among them, six lanostane triterpenoids (19-24,35,36,39,40) and three prenylate phenolics (25-27) were new compounds.3,4-seco-trinorlanostane triterpenoids with a special seven-member B ring (39,40) and prenylate phenolic with three-cycle skeleton (27) were isolated for the first time and their biosynthetic pathway were proposed. We also evaluated their cytotoxic activities and anti HIV-1activities. The results showed that21and31displayed inhibitory against HL-30, MCF-7and SW480cell lines with IC50values10.5,11.3,18.0μM and14.5,16.4,17.3μM, respectively. Meanwhile, others showed weak inhibition.
