Separately treat the cells with Maleicacid rosiglitazone and blood serum of no drugs,coptis chinensis,dried rehmannia root and their combination,setting fat cells with no treatment as the normal group.Collect cellsupernatant and detect its glucose consumption in the method of glucose oxidase.Part two Compare the effect of blood serum of coptis chinensis,driedrehmannia root and their combination on the secreting and mRNAexpressing of adiponectin, leptin and resistin of IR fat cells.Methods of inducing3T3-L1cells to mature fat cells, inducing IR model of fat cellsand setting different groups are all the same as part one. Collect cell supernatant and extractRNA of cells and then compare the effect of blood serum of coptis chinensis,driedrehmannia root and their combination on the secreting and mRNA expressing ofadiponectin, leptin and resistin of IR fat cells in the method of ELISA and RTFQ PCR.ResultsPart one IR model of3T3-L1fat cells and effect of blood serum of coptischinensis,dried rehmannia root and their combination on glucoseconsumption of IR fat cells.Compared with the normal group, the glucose consumption of the model group wasSignificantly reduced(P<0.01); compared with the model group, the glucose consumptionof coptis chinensis group, dried rehmannia root group, combination group andRosiglitazone Maleate group were significantly increased (P<0.01);The glucoseconsumption of combination group was significantly more than either the coptis chinensisgroup or the dried rehmannia root group(P<0.05).Part two The effect of blood serum of coptis chinensis,dried rehmannia rootand their combination on the secreting and mRNA expressing ofadiponectin, leptin and resistin of IR fat cells.Compared with the normal group, the secreting of adiponectin of the model group was significantly reduced(P<0.01), the secreting of leptin and resistin was significantlyincreased(P<0.01); Compared with the model group, the secreting of adiponectin of driedrehmannia root group, combination group and Rosiglitazone Maleate group weresignificantly increased (P<0.01), there was no significant difference between the effect ofcombination group and dried rehmannia root group in the secreting of adiponectin andleptin (P>0.05),but there was in resistin; Compared with the model group, the secreting ofleptin and resistin of dried rehmannia root group, combination group and RosiglitazoneMaleate group were significantly reduced(P<0.05, P<0.01),and the secreting of resistin ofcombination group was significantly lower than either single medicine group(P<0.05).Compared with the normal group, the mRNA expressing of adiponectin of the modelgroup was significantly reduced(P<0.01), the mRNA expressing of resistin wassignificantly increased(P<0.01); compared with the model group, the mRNA expressing ofadiponectin of every group were significantly increased(P<0.05,P<0.01)except the group ofblood serum with no drug, in addition, the mRNA expressing of adiponectin ofcombination group was significantly higher than either coptis chinensis group or driedrehmannia root group(P<0.01,P<0.05);there was no significant difference between theeffect of the model group and any treatment group on the mRNA expressing of resistin(P>0.05).ConclusionCoptis chinensis, dried rehmannia root and their combination are effective onincreasing the secreting or mRNA expressing of adiponectin of IR fat cells to improveinsulin resistance. Coptis chinensis, dried rehmannia root and the combination are effectiveon decreasing the secreting of leptin and resistin. This may be one of the mechanisms thesedrugs improve IR.
