ObjectiveTo investigate the relationships between IL-6and protein expression ofBcl-2, CyclinD1, VEGF and the STAT3pathway in epithelial ovarian cancerSKOV3.Methods1、SKOV3cells were treated for24and48hour with diverse concentrationsof IL-6(0,10,20,40ng/ml)respectively. The proliferation and apoptosis of cellswere detected by MTT and flow cytometry. The mRNA expression of Bcl-2,CyclinD1and VEGF in cells was determined by RT-PCR. The proteinexpression of Bcl-2, CyclinD1, VEGF and p-STAT3in cells was measured bywestern blotting.2、SKOV3cell were treated, in the presence of optimal concentrations ofIL-6(20ng/ml), with diverse concentrations of AG490(0,20,40,80,160μmol/l),a specific JAK kinase inhibitor, respectively. The proliferation and apoptosis ofcells, the mRNA and protein expression of Bcl-2, CyclinD1, VEGF and p-STAT3was remeasured, as previously described.3、The results were expressed as mean±standard error of the mean(SEM). SPSS16.0was involved and statistical analyses were performed byT-test for paired samples comparisons, ANOVA for multiple comparisons.Significance was assumed at P<0.05.Results1、With the increase of IL-6concentration(10,20ng/ml), the action of promoting proliferation(P<0.01)and inhibiting apoptosis(P<0.01)wereenhanced, the mRNA and protein expression of Bcl-2, CyclinD1, VEGF andp-STAT3were increased(P<0.01), compared to the control group. Furthermore, the peak action was obtained at the concentration of20ng/ml. There weredifferences between48h and24h(P<0.01). Compared with IL-6groups of20ng/ml, the group of40ng/ml is no significance(P>0.05).2、With the increase of AG490concentration(20,40,80μmol/l), the actionof inhibiting proliferation(P<0.01)and promoting apoptosis(P<0.01)wereenhanced, and the mRNA and protein expression of Bcl-2, CyclinD1, VEGF andp-STAT3were decreased(P<0.01), compared to the IL-6control group.Further more, the peak action was obtained at the concentration of80μmol/l.Compared with AG490groups of80μmol/l, the group of160μmol/l is nosignificance(P>0.05).ConclusionIL-6promotes the gene transcription and expression of CyclinD1, VEGFand Bcl-2, contributing to the proliferation of ovarian cancer and inhibiting itsapoptosis may via STAT3pathway.
