| Objective:To investigate the correlation between gene polymorphisms of Fn g-ene and renal calcium oxalate stones susceptibility in the Uygur nationality. Meth-od:129 patients with calciu-m oxalate stones (case group) and 94 healthy people (c ontrol group) were enrolled in our study, peripheral blood was collected and geno-mic DNA were extracted from peripheral blood, polymerase chain reaction restricti-on fragment length polymorphism (PCR-RFLP) was performed to analysis polymor-phisms rs6725958, rs10202709 and rs35343655 of Fn, compare association between different genetypes and calcium oxalate stones susceptibility, and made a compreh-ensive analysis of these SNP locies distribution of alleles in Xinjiang Uygur calciu-m oxalate stones patients and healthy people. â‘¡Immunohistochemistry was used to detect the expression of Fn in kidney tissue between the case group and the con-trol group,then make a comprehensive statistical analysis of test results. Results:Th-e results of genetype and alleles of Fn gene reveals that the distributation of three locis goes with Hardy-Weinberg equilibrium,there has no not statistically difference between the three gene sites,Immunohistochemistry results reveals that calcium oxa-late stones group has a higher expression rate, statistically difference was found b-etween t-he two groups. Conclusions: â‘ rs6725958, rs10202709 and rs35343655 p-olymorphisms of Fn gene has no stasitically association with calcium oxalate stone-s susceptibility and may not be the predisposing genes of calcium oxalate stones.â‘¡ Fn protein in renal cortex were significantly different in both groups,suggesting that the some unfound molecular mechanisms of Fn gene to inhibit the formation of c-alcium oxalate stones, our results may related to race and sample size, and still n -eed further study. |
