Objective:To explore Chinese medicine pairs remove "phlegm " and "blood stasis" from "Yiqi ditan zhuyu Formula" in the role of improving local inflammatory microenvironment, removing inflammatory mediators, keeping the balance of cytokines and slow the progression of airway remodeling, seek more targets for medical prevention and treatment of COPD, airway remodeling intervention to provide theoretical basis for the development based on TGF- beta 1 / Smad pathways.Methods:Copy COPD airway remodeling in rats model, observe the change induced fibrosis key fibrosis- TGF- beta 1, were given remove "phlegm" and "blood stasis" Chinese traditional medicine pairs of low, medium and high dose intervention, by ELISA and Western blot method to detect lung tissue of SIgA, plgR/SC, VEGF, MMP-9 and TIMP-1 and TGF-beta 1 / Smad pathways of TGF-beta 1, Smad2/3, Smad7 expression level, explore the medicine "activating blood、removing stasis" improve airway inflammation, slow down the development of the mechanism of airway remodeling.Results:1. HE staining of lung tissue biopsy resultsBronchioles in lungs of rats in the model group around a large number ofinflammatory cell infiltration, airway wall can be seen a large number of fibrous tissue hyperplasia, stenosis, dilated alveolar, alveolar partial integration into bullae,clearly visible manifestations of emphysema, indicating COPD model successfully copied. Chinese traditional medicine pair which can remove "phlegm" and "blood stasis" for the low dose group of lung pathology were improved.2. Each lung tissue in SIgA, plgR/SC, VEGF, MMP-9 and TIMP-1, TGF-beta 1,Smad2/3, Smad7 expressionModel group rats, SIgA, plgR/SC, TIMP-1, VEGF, TGF-beta 1, Smad2/3expression level were significantly increased, MMP-9、Smad7 expression level were significantly decreased, the difference was statistically significant(P< 0.05).Compared with model group, Chinese traditional medicine pairs which can remove "phlegm" and "blood stasis" for the low dose group SIgA, plgR/SC, TIMP-1, VEGF,TGF-beta 1expression level was lowered obviously, has an obvious effect rise in MMP-9, the difference was statistically significant(P < 0.05). Chinese traditional medicine pair which can remove phlegm in low dose group of Smad2/3 expression in rat lung tissue is reduced, the difference was statistically significant(P < 0.05).Chinese traditional medicine pair which can remove blood stasis of low, medium and high dose group of Smad7 expression levels, the difference was statistically significant(P < 0.05).Conclusion:1.Profit lung removing stasis medicine of Chinese traditional medicine pair which can remove phlegm through "removing stasis" intervention of COPD airway remodeling may target one by inhibiting the expression of TGF-beta 1 blocked Smad2/3 phosphorylation.2.Chinese traditional medicine pair which can remove phlegm for regulation of TGF-beta 1/Smad pathway to render certain dose-effect relationship, to clinical equivalent dose adjustment is the most obvious.3.Chinese traditional medicine pairs which can remove "phlegm" and "blood stasis" through by COPD airway remodeling pathogenesis of inflammatory cytokines SIgA, plgR / SC, TIMP-1, MMP-9, VEGF of expression were regulated, slowing airway inflammation, is intervention COPD development of one of the mechanisms ofairway remodeling, clinical equivalent dose is better.
