Objective:To study the rationality and feasibility of the way, which the way to establish the model of syndrome of Yang deficiency was added to the way to establish the model of insomnia model, to establish and evaluate the insomnia rat models under syndrome of Yang deficiency, and to explore the suitable doses of hydrocortisone. To study efficacy of Guizhi-Gancao-Longgu-Muli-Decoction (GGLMD) and its parts in insomnia rats under syndrome of Yang deficiency and research influence of it on content of 5-hydroxy tryptamine (5-HT) and Noradrenaline (NE) in brain of these rats, and then explore possible mechanism of GGLMD treatment.Method:Experiment Ⅰ Establishment and evaluation of the insomnia rat models under syndrome of Yang deficiency40 SD rats were randomly divided into the control group, the insomnia group, the light Hydrocortisone insomnia group, the moderate Hydrocortisone insomnia group and the substantial Hydrocortisone insomnia group. Rats in the control group were given saline in intraperitoneal injection and intramuscular injection. Rats in the insomnia group were given para-chlorophenylalanine (PCPA) in intraperitoneal injection. The remaining groups were given different doses of hydrocortisone in intramuscular injection during 7 days, after that,they were given PCPA in intraperitoneal injection during two days to making the models of rats.Experiment Ⅱ The efficacy of GGLMD and its parts on 5-HT and NE content level in insomnia rats under syndrome of Yang deficiency56 rats were randomly divided into control group (CG), model group (MG), Guizhi-gancao group (GZGC), Longgu-muli group (LGML), Gancao-guizhi group (GCGZ), Guizhi-Gancao-Longgu-Muli-Decoction group (GGLM), clonazepam group (LXDP).Rats in CG were given normal saline in gavage and rats in Other groups ere established insomnia model of Yang Deficiency. Rats in MG were given normal saline in gavage and rats in GZGC, GCGZ, LGML, GGLM were given corresponding decoction of traditional Chinese medicine. Rats in LXDP were given solution of clonazepam. Behavior, daily water intake, tail temperature, r value of image of tail warms and claws and content of 5-HT, NE were observed after five days treatment.Result:Experiment Ⅰ Establishment and evaluation of the insomnia rat models under syndrome of Yang deficiency1. Two rats died in the substantial Hydrocortisone insomnia group, with rats in the other group surviving. Rats in insomnia group showed dark and messy hair, disappeared sleep rhythm, constant motion, bad temper than those of the rats in the control group. All insomnia rats with interference of Hydrocortisone showed dark and messy hair, disappeared sleep rhythm, curled lying, bad temper than those of the rats in the insomnia group.2. With significance level a=0.05, Rats in the moderate Hydrocortisone insomnia group and the substantial Hydrocortisone insomnia group performed lower water intake and tail temperature and lower r value of tails and claws than those of rats in the control group and the insomnia group.3. With significance level a=0.05, Rats in the moderate Hydrocortisone insomnia group and the substantial Hydrocortisone insomnia group performed lower content of 5-HT and higher content of NE than those of rats in the control group and the insomnia group.Experiment II The efficacy of GGLMD and its parts on 5-HT and NE content level in insomnia rats under syndrome of Yang deficiency4. Rats in the GZGC, GCGZ, LGML, GGLM and LXDP, which were awakened more easily than those in CG, showed brighter hair, better sleep rhythm, more sleeping time and less motion time during the day than those in MG.5. With significance level a=0.05, Rats in GGLM and LXDP performed higher water intake and tail temperature and higher r value of tails and claws than those of rats in CG and MG.6. With significance level α=0.05, Rats in GGLM and LXDP performed higher content of 5-HT and lower content of NE than those of rats in CG and MG, while, there were no significant differences of the content of 5-HT and NE between rats in GGLM and LXDP.Conclusion:The way, which the way to establish the model of syndrome of Yang deficiency was added to the way to establish the model of insomnia model is feasible. Suitable dosage of PCPA is 300mg/kg in intraperitoneal injection and suitable dosage of hydrocortisone is 10mg/kg in intramuscular injection in the model establishment. GGLMD perform significantly effect for insomnia of syndrome of Yang deficiency, and its sedative effect, in which the concha ostreae and Longgu perform an important role, is as much as that of LXXP. With the help of cassia twig, which has the effect of yang addition, and licorice, have a better effect on insomnia of this type.
