In this paper, we made a system isolation study on the chemical constituents of Eriosema chinense Vogel. The present article developed a high speed countercurrent chromatography (HSCCC) method for the separation and purification of the chemical constituents from Eriosema chinense Vogel. On the basis of physical-chemical constants and spectral data(UV,IR,MS,1H-NMR and 13C-NMR), the structures of the isolated compounds were identified.We also discussed about the biological activities of the system isolation abstracts from Eriosema chinense Vogel.The main result is formulated in the following points.1. Study on the chemical constituents of petroleum ether extractSeparation of petroleum ether extract by HSCCC was studied in this paper.The solvent system N-hexane/ Ethyl Acetate/Methanol/water(5:5:7:3,V/V) was used,at a flow rate of 2.0 mL·min-1 while the apparatus rotated at 700 r·min-1, and the effluent was detected at 254 nm. Two compounds were separated firstly from Eriosema chinense Vogel and were new compouds. The molecular formulas of the compound 1 and compound 2 are C26H28O6 and C26H30O6. Through preliminary identification , the compound 1 is 3-Hydroxy-2-(4-Hydroxy-phenyl)-5-methoxy-8,8-dimethyl-10-(3- methyl-but-2-enly)-2,3-dihydro-8H-pyrano[3.2-g]chromen-4-one and the compound 2 is 6,8-(3-methyl-but-2-enly) -5-methoxy-3,7-trihydroxydihydroflavone. The concrete structure of two compounds still needs further study.2. Study on the chemical constituents of ethyl acetate extractThe ethyl acetate extract from Eriosema chinense Vogel was seperated by preparative high speed countercurrent chromatography.The solvent system N-hexane/Ethyl Acetate/Methanol/Water(1:0.6:1:0.6,V/V) and N-hexane/Ethyl Acetate/Methanol/water(3:6:3:6,V/V)were used at a flow rate of 2.0 mL·min-1,while the apparatus rotated at 700 r·min-1 and the effluent was detected at 254 nm.Three compouds were obtained from the fraction of ethyl acetate.Up to now, three compounds which were isolated from this plant for the first time were identified as genistein,kaempferol and 4’-Methoxy-5,7-dihydroxy isoflavone. 3. Experimental study on biological activity of extractives from Eriosema chinense VogelThe results showed that three kinds of extractives had different bacteriostatic activity to Staphylococcus aureus.The mice ear edema caused by xylene were significantly suppressed by three kinds of extractives. The cough-settling and phlegm-dispelling effects were observed on the ammonia water-induced cough model of mice and phenol red expectoration method. The results showed that three kinds of extractives had significant antibechic effects in mice and could increase phenol red output of trachea in mice expect ethyl acetate extractⅠ.
