| At present,the population of fatigue sub-health was numerous in our country,which had become a major problem threatening human health.In process of looking for ways to improve this problem,concentrated blueberry juice had many natural active ingredients with stronger health care effect,and made up for the defect of chemicals such as methylphenidate producing side effects to the body.This study evaluated the effect of the concentrated blueberry juice on anti-fatigue and survey the mechanism involved preliminarily.The main research contents and results were as follow:(1)A stable model of fatigue in mice was established through a certain intensity of forced swimming in mice and regular testing index of fatigue.Results showed that,after two weeks of forced swimming with 25 min in constant temperature water at 25±1 °C everyday,behavior characteristics of the male mice of 15~20 g weight was changed.At the same time,T-SOD activity and LDH activity were decreased,and metabolic accumulation such as MDA and BUN levels were increased.The mice was continued to maintain the same exercise intensity in a week,and their fatigue status did not recover.It proved that two weeks of forced swimming could be used to establish a stable model of the fatigue.(2)The mice were prescreened and forced swimming training.Then choosing the physiological saline as control group,and choosing the high and low dose of concentrated blueberry juice and VC as the experimental groups separately.After the processing of dosing and swimming training,the behavioral characteristics of each group mice was observed and the swimming exhaustion time was measured.The changes of T-SOD activity,MDA content and LDH activity in the plasma and the brain tissue homogenate protein were tested.The changes of BUN content in the plasma were also tested.The method of HE stain was performed to observe the tissue pathological change of mice.Results showed that,compared with the control group of swimming,high dose of concentrated blueberry juice could improve the behavioral characteristics of fatigue mice obviously and prolong swimming exhaustion time.It could improve T-SOD and LDH activity significantly,reduce the production of MDA of the plasma and the brain tissue homogenate.Besides it could reduce the production of BUN of the plasma.It also could prevent the formation of cell clearance.Compared with the high dose concentrated blueberry juice,the improved effect of low dose concentrated blueberry juice and VC treatment group was not obvious.(3)The mixed culture of the original generation nerve cells were obtained from neonatal mice,then the astrocytes were purified and the purity were more than 95%.Using H2O2 to simulate the hypoxia environment in vivo induced by exercise,and exploring the relationship between the anti-fatigue activity of concentrated blueberry juice and mechanism of oxidative stress.Results showed that,concentrated blueberry juice could significantly improve injury of original generation astrocytes on the morphology and quantity induced by H2O2,and reduce the Nrf-2 protein in endochylema to protect the cells.The above results showed that a certain intensity of forced swimming could establish a stable model of fatigue and concentrated blueberry juice treatment could obviously improve the fatigue sub-health state in mice,of which mechanism might be related to the Nrf-2 protein regulating the antioxidant pathway. |
