Objective: analysis of IDH1 mutation,MGMT and Ki-67 expression in glioma was performed to evaluate the prognosis of patients..Methods: Immunohistochemistry was used to detect the expression of IDH1,MGMT and Ki-67 in 110 patients with glioma.Long-term follow-up was conducted to analyze the correlation between the expression of these three molecular markers and prognosis. Results: The total mutation rate of IDH1 in 110 gliomas was 33.64%,and the mutation rates of IDH1 in WHO II,WHO III and WHO IV were 55.55%,32% and 10%,respectively.Ki-67 was positive in gliomas of different grades.Ki-67 expression in high-grade gliomas was significantly higher than that in low-grade gliomas. The rate of high expression of Ki-67 in IDH1 mutant group and IDH1 wild type group was 8.11% and 12.99%,respectively.The rate of low expression of Ki-67 in IDH1 mutant group and IDH1 wild type group was 78.38% and 55.84%,respectively.The median survival time of MGMT methylation was significantly higher than that of non-methylation patients.Low-grade gliomas mainly involve the frontal and temporal lobes. With the increase of glioma grade, the involvement of parietal and occipital lobes gradually increases. The ratio of Cho/Cr increased with the increase of Ki-67 expression. Conclusion: The glioma patients with IDH-1 mutation,MGMT methylation and low expression of Ki-67 had lower pathological grade and better prognosis, and there was a certain correlation between them.Low-grade gliomas mainly involve the frontal and temporal lobes. With the increase of glioma grade, the involvement of parietal and occipital lobes gradually increases. The ratio of Cho/Cr increased with the increase of Ki-67 expression.
