Research purposes:Expression of IDH1 mutation,MGMT promoter methylation and TERT mutation in high-grade gliomas,and to analyze its impact on the outcome of the high-grade gliomas Methods:In First Affiliated Hospital of Anhui Medical University,choose from September 2017 to December 2018 pathologically confirmed high-grade glioma samples using molecular detection methods clear sample IDH1 mutation,MGMT methylation and TERT mutation,with SPSS22.0 line data processing,through c2 test to determine the correlation among IDH1 mutation ?MGMT methylation and TERT mutation,using Kaplan-Meier analysis IDH1 mutation,MGMT promoter methylation and mutations in TERT of high-grade gliomas prognosis,and analyze the impact of age,WHO pathological grade,IDH1 mutation,MGMT promoter methylation,TERT mutations and other prognostic factors for high-grade gliomas using COX model.Research result : There are correlations between IDH1 mutation and MGMT methylation,between IDH1 mutation and TERT mutation.MGMT methylation was no significant correlation with TERT mutation.By survival analysis,high-grade gliomas IDHl mutation better prognosis(P <0.001),MGMT methylation in overall survival in high-grade gliomas unmethylated MGMT significantly longer than(P <0.001),TERT promoter mutations after poor prognosis in high-grade gliomas(P <0.001),in the packet analysis,IDHl wild-type/MGMT unmethylated/TERT wild-type of high-grade gliomas had worst prognosis.Analysis conclusion: IDH1 mutation,MGMT methylation and TERT mutation are prognostic factors of high-grade glioma,combination of the three makes is more sense to judge the high-grade glioma outcomes.
