ObjectiveIDH1mutations and MGMT expression,in different grade gliomas and normal brain tissue by immunohistochemistry (immunohistochemistry technique IHC) method, research, to discuss the IDH1gene mutations associated with MGMT expression in order to further explore glialthe pathogenesis of the tumor, the results provide some basis for clinical diagnosis and treatment of glioma.MethodsUsing immunohistochemical techniques were used to detect IDH1mutations in51cases of grade gliomas and10cases of normal brain tissue and MGMT expression, and correlation analysis.Results1.IDH1mutations in51cases of glioma patients in26cases of positive expression of its overall positive expression rate of50.98%. Of I-III grade glioma IDH1mutations in the expression of an upward trend, the expression of IDH1mutations in IV grade glioma decline expressed by the IDH1mutations between the different levels of statistical analysis of glioma was no significant difference (P>0.05).2.51cases of glioma patients with MGMT positive expression in28cases, its overall positive expression rate of54.90%. MGMT expression rate with elevated glioma pathological level the overall upward trend, not statistically significant (P>0.05) by the difference between the different levels of statistical analysis of glioma.3.10cases of decompression surgery in hypertensive intracerebral hemorrhage line, the removal of the normal brain tissue specimens IDH1mutations and MGMT expression was negative or weakly positive, the IDH1gene mutations in gliomas and MGMT expression there is a significant difference (P <0.05).4.51cases of glioma tissues of patients with IDH1mutations and MGMT expression was associated with gender, age differences were not statistically significant.Conclusions1.Different grade gliomas IDH1mutations expression rates, IDH1mutations I III grade glioma express an upward trend, the expression of IDH1mutations in IV grade glioma decline, but the statistical analysisthe difference was not statistically significant, the expression of IDH1mutations had no correlation with tumor grade.2. Different levels of glioma MGMT expression is not the same as gliomas increased levels of MGMT expression rate the overall upward trend, gliomas, the higher the level of MGMT expression is a relatively high rate, but the statistical analysis of the difference not statistically significant, indicating that MGMT expression correlation with tumor grade.3. The correlation between IDH1mutation and MGMT expression in gliomas, suggesting the occurrence of both glioma, the development may have the role of mutual adjustment, the specific mechanism of action to be studied. Joint IDH1mutation and MGMT detection to determine the prognosis of glioma patients and the development of rational cancer chemotherapy.4. IDH1mutations and MGMT expression was negative or weakly positive in normal brain tissue.
