Objective: To investigate the relationship between the expression of miRNAs in human gliomas with epilepsy and pathological grade;and the relationship between the expression of IDH1 mutation,MGMT,Ki67 protein and the pathological grade and prognosis of glioma,and to discuss the relationship between miR-128,miR-31,miR-497 and IDH1 mutant,MGMT,Ki67.Methods: The surgical specimens of 21 cases of glioma,13 cases of low-grade glioma(7 cases with history of epilepsy before operation,6 cases without history of epilepsy),8 cases of high-grade glioma(no history of preoperative epilepsy)and 3 cases of brain trauma were collected.quantitative Real Time Polymerase Chain Reaction was used to detect the expression of miR-128,miR-31 and miR-497 in surgical specimens of glioma patients.Immunohistochemical method was used to detect the expression of IDH1 mutation,MGMT and Ki67 protein.Real-time PCR was used to detect the expression of miR-128,miR-31 and miR-497 in glioma cell line U87 and human microglial cell line HMC3.Results:(1)The expression level of miR-128 in patients with low-grade glioma with a history of epilepsy before operation was significantly higher than that in patients without epilepsy.(2)The expression level of miR-128,miR-31 and miR-497 in low-grade gliomas was significantly higher than that in high-grade gliomas,and significantly lower than that in normal brain tissues in gliomas,while the expression level in glioma cell line U87 was significantly lower than that in human microglial cell line HMC3.(3)Survival curve analysis showed that the survival time of IDH1 mutation group was significantly longer than that of wild type group,MGMT negative expression group compared with positive group and Ki67?10%group compared with Ki67>10% group.(4)The expression levels of miR-128,miR-31 and miR-497 in IDH1 mutation group were significantly higher than those in wild type group;The expression level of miR-128,miR-31 and miR-497 in MGMT negative expression group was significantly higher than that in positive expression group;and the expression level of miR-128,miR-31 and miR-497 in Ki67?10% group was higher than Ki67>10% group.Conclusion:(1)miR-128 is a potential target for the treatment of gliomas and patients with epilepsy.(2)The expression levels of miR-128,miR-31 and miR-497 were negatively correlated with the pathological grade of gliomas,and the upward adjustment predicted longer survival time and better prognosis.(3)IDH1 mutation,MGMT negative expression,Ki67?10% or lower indicate longer survival time and better prognosis.
