| Objective:By observing the kidney and strong bone pill of spleen and kidney Yang deficiency type osteoporosis rats normally,body quality and anus temperature,viscera index,bone mineral density,bone biomechanics,bone transformation indexes and the change of the vertebral body HE staining results,study the kidney and strong bone pill of spleen and kidney Yang deficiency type osteoporosis prevention and cure function,provide experimental basis for its widely clinical application.Methods:Experiment 1:24,SPF female SD rats were randomly divided into blank control group,model group and control group,in addition to the blank control group,model group with reference to the literature by castration rats(removal of the ovaries)+injection of hydrocortisone dishonor+senna lavage+diet weary excessively method,establishing the spleen kidney Yang deficiency type osteoporosis rats model,control exposure after bilateral ovaries do not removed.Urine D-xylose excretion rate,serum bone formation and bone resorption markers,bone density and bone biomechanics,organ index,histophistological examination and other indexes of rats in the three groups were determined to evaluate the feasibility and success rate of the modeling method.Experiment 2:56 SPF female SD rats were randomly divided into blank control group,sham operation group and model group.Except blank control group,rats in other groups were treated with the following methods:castration(resection of bilateral ovaries of female SD rats)+injection of hydrocortisone+senna leaf gavage+diet loss+excessive fatigue,and model replication was conducted.In the sham operation group,only bilateral ovaries were exposed without excision.The model was duplicated for 8 weeks.After the success of osteoporosis animal model,each group was divided into Wenshen Qianggu Pill high-dose,medium-dose and low-dose groups and western medicine groups,with 8 rats in each group.The blank control group and sham operation group were given equal dose of normal saline by gavage.Western medicine group was given alendronate sodium aqueous solution by gavage.4 times a week for 8 weeks.The general condition,body weight and anal temperature of rats were observed and recorded.The indexes of spleen,kidney,uterus and other organs of rats were detected.Bone density and bone biomechanics of rats were detected.Bone transformation indexes such as TRAP,U-NTX,BALP,E2,U-CTX and BGP in serum were detected.He staining results of vertebral bodies were observed and analyzed.Results:Experiment 1:Compared with blank control group,the excretion rate of D-xylose in urine of rats in model group was significantly decreased(P<0.05);In the serum bone metabolism indexes,the bone formation promoting markers BALP and E2 were significantly decreased(P<0.05),while the bone formation inhibiting markers BGP,TRAP,U-NTX and U-CTX were significantly increased(P<0.05).Bone mineral density and bone biomechanics were significantly decreased(P<0.05);Bone trabeculae decreased;In addition,the model rats showed obvious symptoms of spleen-kidney Yang deficiency,such as chills and loose stool.Experiment 2:Wenshen Qianggu Pill could significantly improve the general state of model rats,increase the amount of diet,increase body weight,anal temperature,and significantly relieve the symptoms of spleen-kidney Yang deficiency such as chills and loose stools.The BMD of femur and spine of rats in model group was increased(P<0.05),and bone biomechanics was enhanced(P<0.05).The serum levels of BALP and E2 were increased(P<0.05),and the serum levels of BGP,TRAP,U-NTX and U-CTX were decreased(P<0.05);The percentage of cortical bone area,average trabecular thickness,trabecular area and trabecular volume were increased(P<0.05).Conclusion:Experiment 1:The method of castrating(rat ovary excision)+hydrocortisone injection+sennata leaf gavage+diet loss+overfatigue combined with the method of building rat model was feasible,and the model method was more close to the pathogenic factors of human spleen-kidney Yang deficiency type osteoporosis.Experiment 2:the kidney and strong bone pill can obviously improve the symptoms of spleen and kidney Yang deficiency rats,increase the rat bone mineral density,bone biomechanics,elevated serum BALP,E2 levels,reduce serum BGP,TRAP,U-NTX,U-CTX level,increase the cortical bone area percentage and average thickness and the trabecular bone trabecular bone area,trabecular bone volume,and treatment of spleen and kidney Yang deficiency type osteoporosis. |
