The Variation Of Inflammatory Factors Related To The Treatment Of Yangming Disease(yangming Fu-viscera Excess Syndrome)and Experimental Study On The Intervention Effect Of Dacheng Qi Decoction

Objective:To investigate the changes of tumor necrosis factor(TNF-α),interleukin-6(IL-6)and Nuclear transcription factor-κB(NF-κB)of intestinal mucosa-related inflammatory factors in the treatment of Yangming disease(Yangmingfu-viscera excess syndrome)and after the intervention of Dachengqi Decoction.Methods:A total of 290 healthy non-pregnant SD rats were randomly divided into normal group(n=90),model group(n=100)and treatment group(n=100).The normal group was given the same amount of distilled water every day,and the model group was given the model of Yangming disease(Yangming fu-viscera excess syndrome)to experimental rats by injecting the traditional Chinese medicine heat medicine,self-fecal suspension and liposolysaccharide.After the successful modeling,the treatment group were given Dachengqi decoction for intervention at 5:00-7:00 and17:00-19:00,and observed for 5 consecutive days.After the completion of model making and drug intervention,9 rats were extracted from normal group,10 rats were extracted from model group and treatment group every day,and tissue homogenates were obtained after head amputation to observe the changes of tumor necrosis factor(TNF-α),interleukin-6(IL-6)and nuclear transcription factor-κB(NF-κB)levels.Results:Changes of TNF-α : TNF-α expression in normal group was lower than that in model group and treatment group(P<0.01).The expression of TNF-α in the model group showed a rhythm,which reached the peak on the second day,decreased to the lowest on the third day,and then tended to be stable.The expression of TNF-αin the treatment group increased briefly at "recovery time" and then decreased gradually,which was lower than that at "non-recovery time"(P<0.05)and finally stabilized.Changes of IL-6: The overall trend of IL-6 expression in the model group was higher than that in the normal group.On day 4,the expression of IL-6 in the model group was higher than that in the normal group(P<0.05),the "non-recovery time" of the model group on day 5 was lower than that of the normal group(P<0.05).The expression of IL-6 in the model group all reached the peak on the first day,and then gradually decreased and became stable.After drug intervention,the expression of IL-6 in the treatment group was higher than that in the normal group and the model group(P<0.05).The level of IL-6 in the treatment group was higher than that in the "non-recovery time" on the first day,and lower than that in the "non-recovery time" on the third and fifth days,showing significant differences(P<0.05).Changes of NF-κB: The overall expression of NF-κB in the model group was higher than that in the normal group,and the expression of NF-κB in the model group was gradually increased,and reached the peak on the 3rd day of recovery time,and then decreased gradually.After drug intervention,the content of NF-κB in the treatment group was lower than that in the model group(P<0.05).The content of NF-κB in the treatment group at the 1st,4th and 5th day was lower than that at the non-recovery time(P<0.05).Conclusion:On the whole,the expression of TNF-α,IL-6 and NF-κB in the model group was higher than that in the normal group,and the content expression of the three indexes in the model group showed a rhythm,in which TNF-α and IL-6 were relatively fast,and NF-κB was slightly lagged.After the intervention of Dachengqi Decoction,the expressions of TNF-α,IL-6 and NF-κB in the treatment group began to increase briefly,and then gradually decreased and tended to be stable.With the progression of disease,the levels of TNF-α,IL-6 and NF-κB in the "recovery time" were significantly decreased compared with those in the "non-recovery time".
