ObjectivePostherpetic neuralgia is a common neuropathic pain,its incidence is increasing year by year,but the current treatment effect is not good.Traditional Chinese medicine(TCM)has a long history of understanding herpes zoster,but it is difficult to comprehensively and systematically understand the multi-target pharmacological mechanism of action of TCM due to its various ingredient.With the emergence of bioinformatics,it is possible to explore the mechanism of action of TCM compounds.In this paper,data mining was used to analyze the medicaion rules of TCM in the treatment of postherpetic neuralgia,and core drugs was extracted and explored the mechanism of action of traditional Chinese medicine with network pharmacology.MethodsThis article took "postherpetic neuralgia" as the object,and all published literatures about TCM treatment of postherpetic neuralgia were obtained on the Chinese Journal Database(CNKI),Wanfang Data and the Chongqing VIP Chinese Science and Technology Periodical Database.The prescription data set of traditional Chinese medicine for the treatment of postherpetic neuralgia was obtained according to certain standard of absorption and discharge.Then,the traditional Chinese medicine data set was normalized with terms,and the properties of frequency,four qi,five flavors and meridian return of traditional Chinese medicine were counted.Combined with association rule algorithm,the core formula structure and formula rule were analyzed,and the core prescription was obtained.Active ingredients of the core prescription were retrieved from the BATMAN-TCM database,and a matching score(Score >20,P <0.05)was set through the target prediction system equipped by the platform itself,which was used as a screening condition to collect the predicted targets corresponding to active ingredients.Disease targets were collected in Gene Cards database using "neuropathic pain" as the search term.The predicted targets of the core prescription for the treatment of neuralgia were mapped to the intersection of the predicted targets of the core prescription for the treatment of neuralgia.The predicted targets were imported into the String database to obtain the interaction relationship between the predicted targets.Cytoscape was used to build a network model,and the GO enrichment analysis and KEGG pathway enrichment analysis was conducted.ResultsA total of 212 prescriptions were included in this study.angelica,licorice,rhizoma Corydalis,safflower and bupleurum were the most frequently used drugs of the first five flavors.The main tradition Chinese medicine treating postherpetic neuralgia belongs to the warm and cold drugs and the five flavors were mainly bitter,pungent and sweet.Association rule analysis showed that under the condition of 30% support and 0.8 confidence,30 high-frequency TCM combinations were obtained,involving 12 TCM herbs.Combining with the data mining results,the core formula was formulated to be composed of 10 TCM herbs,including Astragalus membranaceus,Bupleurum bupleurum,white peony root,angelica sinensis,safflower,licorice root,rhizoma chuanxiong,rhizoma corydalis,red peony root,peach kernel.10 herbal medicines were used for network pharmacological analysis,and 260 active ingredients were obtained,265 potential targets were obtained.The ingredient target map was constructed through Cytoscape3.5.3,and 265 core prescriptions related to neuralgia were queried through the string database.The interaction diagram between targets,49 core targets were selected.After GO enrichment analysis and KEGG pathway enrichment analysis,15 major pathways were obtained.ConclusionThe core prescription components of traditional Chinese medicine in the treatment of postherpetic neuralgia are: Astragalus membranaceus,Bupleurum bupleurum,white peony root,angelica sinensis,safflower,licorice root,rhizoma chuanxiong,rhizoma corydalis,red peony root,peach kernel.The main targets include a series of inflammatory cytokine-related targets such as IL6,TNF,IL1 B,IL10,TLR4 and IL4.Nerve growth factor-related targets such as BDNF and NGF were also included.The possible pharmacological mechanisms involve the regulation of inflammatory response,anti-oxidative stress,and changes in neuroplasticity.The main pathways include tumor necrosis factor signaling pathway,HIF-1 signaling pathway,PI3K-Akt signaling pathway,cytokine-cytokine receptor interaction,T cell receptor signaling pathway,Toll-like receptor signaling pathway,etc.This reflects the multi-target and multi-channel therapeutic effect of traditional Chinese medicine.
