1.Data mining of traditional Chinese medicine in the treatment of migraine with hyperactivity of liver YangResearch objective:Based on data mining,this paper analyzes the law of traditional Chinese medicine and the composition of core drugs in the treatment of migraine with hyperactivity of liver yang.Research methods:The databases such as China journal full text database(CNKI),Wanfang Data knowledge service platform(WF),VIP and CBM were searched.The search time was from the establishment of the database to November 2021.The clinical literature on the treatment of migraine patients with hyperactivity of liver Yang by Chinese medicine was collected,and the database was established by Excel software,The ancient and modern medical records cloud platform software(personal version v2.3)was used to perform frequency analysis,cluster analysis and association analysis on the drugs extracted from the database.Research results:103 literatures were included in this study,including 128 kinds of traditional Chinese medicine,and the total frequency of drug occurrence was 1145,including Gastrodia elata,uncaria,Ligusticum chuanxiong,Achyranthes bidentata,cassia,white peony,Scutellaria baicalensis,gardenia,mulberry parasitism,Polygonum multiflorum,motherwort,licorice,Eucommia ulmoides,Angelica dahurica,Poria,angelica,Vitex,Rehmannia glutinosa,chrysanthemum,scorpion,bupleurum,earthworm,Tribulus terrestris The medication frequency of Salvia miltiorrhiza ranked the top 24.The total frequency of four Qi in traditional Chinese medicine is 1093 times,ranking from high to low,which are flat medicine,cold medicine,warm medicine,slightly cold medicine,cool medicine,slightly warm medicine,hot medicine,great cold medicine and great heat medicine.The total frequency of five flavors of traditional Chinese medicine is 1620,ranking from high to low as sweet medicine,bitter medicine,pungent medicine,sour medicine,salty medicine,light medicine,slightly bitter medicine,astringent medicine,slightly sweet medicine and slightly pungent medicine.The total frequency of meridians of traditional Chinese medicine is 2265,ranking from high to low: liver,heart,spleen,lung,kidney,stomach,pericardium,gallbladder,bladder,triple energizer,large intestine and small intestine.The results of cluster analysis of traditional Chinese medicine show that traditional Chinese medicine can be divided into two categories: Ligusticum chuanxiong-white peony-Vitex-Angelica-Rehmannia glutinosa-licorice-Angelica dahurica-chrysanthemum,and the other is Achyranthes bidentata-Cassia-Gastrodia-Uncaria-Scutellaria-Gardenia-Eucommia ulmoides-Poria-mulberry parasitism-shouwuteng-motherwort.The results of correlation analysis of traditional Chinese medicine showed that Gastrodia elata and Uncaria are the common drug pairs for the treatment of migraine with hyperactivity of liver yang.The other common drug pairs are shijieming Gastrodia elata,shijieming uncaria,Achyranthes bidentata,shijieming Achyranthes bidentata,Chuanxiong Gastrodia elata,Scutellaria baicalensis gastrodia,etc.Research conclusion:Based on the research results of data mining,it is concluded that Gastrodia elata,uncaria,Ligusticum chuanxiong,Achyranthes bidentata,stone cassia,Radix Paeoniae Alba,Radix Scutellariae and other drugs are mainly used in the treatment of migraine with hyperactivity of liver Yang,the four Qi are mainly flat,cold and warm,and the five flavors are mainly sweet,bitter and pungent,mostly from the three Zang organs of liver,heart and spleen.2.Research on the action mechanism of core drugs based on network pharmacologyResearch objective:Based on the research of network pharmacology,this paper analyzes the core target and mechanism of traditional Chinese medicine in the treatment of migraine with hyperactivity of liver yang.Research methods:By searching tcmsp and tcmid databases,the effective chemical components and related targets of core traditional Chinese medicine were obtained.The disease-related targets were obtained by searching and screening in the disgenet disease database.The Uni Prot protein database was used to annotate the chemical composition targets of the core traditional Chinese medicine,and the genes matching the disease targets were obtained.The corresponding drug effective chemical component target network diagram and core drug key chemical component intersection target disease network diagram were constructed by Cytoscape 3.9.0 software.Then the intersection targets of drugs and diseases are introduced into string 11.5 online platform to construct the protein-protein interaction network diagram,and the obtained protein-protein interaction network diagram is topologically analyzed by using Cytoscape 3.9.0 software to obtain the key targets.Finally,go enrichment analysis and KEGG pathway enrichment analysis were carried out for the intersection targets of drugs and diseases by using R software and related installation packages.Research results:(1)There are 106 effective chemical components of core traditional Chinese medicine(33 species of uncaria,45 species of Gastrodia elata,7 species of Ligusticum chuanxiong and 20 species of Achyranthes bidentata).(2)There are 1926 targets of effective chemical components of core traditional Chinese medicine(764 uncaria,714 Gastrodia elata,35 Ligusticum chuanxiong and 413 Achyranthes bidentata).(3)Disease targets: a total of 742 migraine related targets were obtained.(4)Core drugs-key chemical components-intersection targets-disease network diagram: there are 97 nodes(4 core drug names,41 key chemical components,51 intersection targets,1 disease name)and 279 edges in the diagram.(4)Key chemical components: citric acid,quercetin,wogonin.(5)Key targets: AKT1,TP53,ESR1,CASP8,KRAS.(6)The results of go enrichment analysis showed that a total of 36 go enrichment analysis molecular functions were obtained.The main molecular functions include endopeptidase activity,protease binding,sulfur compound binding,ubiquitin protein ligase binding and so on.The results of go enrichment analysis showed that a total of 860 go enrichment analysis biological processes were obtained,including gland development,response to metal ions,epithelial cell proliferation,response to peptide hormones,etc.The results of go enrichment analysis showed that a total of 20 go enrichment analysis cells were obtained.The main cell components include cell top,membrane raft,membrane micro region,membrane region,etc.(7)The results of KEGG pathway enrichment analysis show that 82 pathways are obtained,mainly involving PI3K-Akt signal pathway,MAPK signal pathway,etc.Research conclusion:Based on network pharmacology research,the key chemical components of core traditional Chinese medicine combination to improve migraine with hyperactivity of liver yang are citric acid,quercetin,wogonin and so on.The key targets are AKT1,TP53,ESR1,CASP8,KRAS,etc.The cell composition of gene products of core traditional Chinese medicine combination chemical components is mainly in cell top,membrane raft,membrane micro region,membrane region and so on.It mainly regulates many molecular functions such as endopeptidase activity,protease binding,sulfur compound binding and ubiquitin protein ligase binding,regulates a variety of biological processes such as gland development,response to metal ions,epithelial cell proliferation and response to peptide hormones,and regulates signal pathways such as PI3K-Akt signal pathway and MAPK signal pathway,so as to treat migraine with hyperactivity of liver yang.
