| Objectives:1.The data mining technology was used to analyze the medication rules and core drugs of the tutor in the treatment of Behcet’s disease heat and toxicity syndrome,and the clinical experience of the tutor in the treatment of the disease was summarized.2.To study the mechanism of action of the core drugs in the treatment of Behcet’s disease heat and toxicity syndrome by the network pharmacological method.Methods:1.To collect the first diagnosis cases of 101 cases of Behcet’s disease heat and toxicity syndrome treated by the tutor in the First Affiliated Hospital of Heilongjiang University of Chinese Medicine from October 2020 to October 2022.The information of outpatient cases and prescription drugs were standardized processed,a database was established and uploaded to the V3.0software of TCM inheritance computing platform.Frequency analysis,association rule analysis and cluster analysis were conducted on the case data.To explore the medication rules and core drugs of the tutor in the treatment of Behcet’s disease,and summarize the treatment experience of the tutor.2.Network pharmacological analysis was conducted on the core drugs used by the tutor for the treatment of the heat and toxicity syndrome of Becet’s disease,and the chemical components and targets of the core drugs were obtained through TCMSP and Batmand-TCM databases.Disease targets were obtained through Gene Cards,Drugbank,OMIM and TTD.The network of Drug,Active Ingredient and Key Target was constructed by Cytoscape software,and 10 key targets and important components were selected.PPI network is constructed by STRING database,and visual analysis is carried out.GO enrichment analysis and KEGG pathway analysis were performed using DAVID6.8 database.Results:1.Among the 101 cases included in this study,58 females accounted for57.43% and 43 males accounted for 42.57%.The number of female patients was higher than that of male patients,and the number of patients in the age group 30-39 was the largest,accounting for 37.62%.2.Among the 101 patients with incandescent syndrome of Behcet’s disease in this study,a total of 31 symptoms were collected,of which 16 had frequency ≥25.The top 10 symptoms were oral ulcers,red,swollen and painful facial choices,insomnia,dry stools,erythema nodules of the skin,dry eyes,joint pain,redness,restlessness and vulvar ulcers.3.A total of 161 kinds of traditional Chinese medicine were counted in this study,among which 22 kinds of traditional Chinese medicine were used more than or equal to 30 times,and the top nine kinds of traditional Chinese medicine were Gentian,gardenia,alisma,sanguinis,wood butterflies,moutan bark,mulberry bark,cassia seed and scutellaria.In the classification of drug efficacy,the top five were clearing heat,improving water and dampness,tonifying deficiency,stopping bleeding,reducing phlegm and relieving cough and asthma.The frequency of four drugs from high to low was cold drugs,flat drugs,warm drugs,cool drugs,heat drugs.The five tastes of medicine from high to low are bitter,sweet,hot,salty and sour.The drug channel from high to low is liver meridian,lung meridian,heart meridian,stomach meridian,spleen meridian,kidney meridian,large intestine meridian,gallbladder meridian,small intestine meridian,three jiao meridian,pericardium meridian.4.Analysis of drug association rules showed 38 drug patterns and 61 drug association rules,and there were 9 core drugs: Gentian,gardenia,alisma,sanguinis,papillon,moutan bark,mulberry bark,cassia seed and scutellaria.Commonly used drug combinations are: Gentian,gardenia;Gentian,alisma alisma;Elm,wood butterfly;Moutan bark,mulberry bark.5.Six groups of drug core combinations were obtained by drug cluster analysis.The core combinations were 1: Sanguinis ulmus,Chrysalis,Gentian,senna,gardenia,cassia,mulberry bark,Moutan bark,and rigidus mori.Core group 2: Gentian,Gardenia,alisma,Moutan bark,mulberry bark,Cassia seed,wood butterfly,Sanguinis sanguinis,scutellaria;Core group 3: Alisma alisma,peony bark,sanguinis ulmus,Gentian,gardenia,mulberry bark,wood butterfly,zombie silkworm,poria;Core assemblage 4: Sanguisorba sanguinalis,Wood butterfly,Gardenia,Gentian,alisma orientalis,Moutan bark,Cassia seed,Septicaria septemba,Dioscorea septemba;Core Group 5: Forsythia,wood butterfly,Cassia,gardenia,sanguinis,Gentian,alisma,poria,atractylodes;Core Group 6: Gardenia,Gentian,alisma alisma,areca nut,Moutan bark,sanguinis ulmus,mulberry bark,Scutellaria scutellaria,grass fruit.6.A total of 97 active ingredients of core drugs were obtained by network pharmacological analysis,with 922 targets,2063 disease targets and 159 intersection targets.The main active ingredients were quercetin,kaempferol,β-sitosterol,Han baicalin,etc.The key targets were TNF,IL-6,AKT1,IL-1β,VEGFA,etc.There were 380 biological function items in GO analysis,involving 294 biological processes,35 cell components and 51 molecular functions.A total of 106 pathways were obtained by KEGG pathway enrichment analysis,mainly involving AGE-RAGE signaling pathway,IL-17 signaling pathway,TNF signaling pathway,PI3K/AKT signaling pathway,etc.Conclusions:1.Among the 101 cases in this study,the ratio of male to female was equal.The main symptoms were oral ulcer,facial redness,swelling and pain,insomnia,dry stool and erythema nodules.2.The main drugs used by the tutor in the treatment of Behcet’s disease syndrome were heat-clearing drugs,moistening drugs,tonifying drugs,hemostatic drugs,phlegm-reducing drugs for relieving cough and antiasthmatic drugs.The four qi were mainly cold and calm,the five were bitter,sweet and occultic,and the main channels were liver channels,lung channels,heart channels and stomach channels.3.The core drugs used by the teacher in the treatment of the heat and toxicity syndrome of Behcet’s disease were Gentian,gardenia,diarrhea,sanguinis,wood chrysalis,mupan bark,mulberry bark,cassia seed,scutellaria scutellaria,and 4 groups of commonly used drug combinations and 6 groups of drug core combinations were obtained.4.Through network pharmacological studies,it was found that quercetin,kaempferol,β-sitosterol,han baicalein and other main components of core drugs may act on multiple targets including TNF,IL-6,AKT1,IL-1β,VEGFA,etc.Behset’s disease may be treated by regulating AGE-RAGE signaling pathway,IL-17 signaling pathway,TNF signaling pathway,PI3K/AKT signaling pathway and other pathways. |
