Objective: The purpose of this study is to explore in depth Professor Tong Ying’s approach,drug characteristics,and core treatment protocols for the treatment of gouty arthritis through data mining and network pharmacology analysis.Methods:1.In order to obtain valid case data,GA patients who attended the clinic of Prof.Tong Ying at the First Clinic of Heilongjiang University of Traditional Chinese Medicine Services between March 01,2019 and December 31,2022 were selected,screened,summarized and organized,and finally entered into an Excel sheet in order to establish a complete database.By utilizing the advanced technology provided by the TCM heritage computing platform,the methods,drug properties and core prescriptions of Professor Tong Ying’s treatment of GA were explored in depth in order to obtain more accurate results.2.Through network pharmacology data analysis,we found that Professor Tong Ying’s core prescriptions for GA contain active ingredients and potential protein targets.We used the TCMSP database system and the Gene Cards and OMIM database systems to examine them to identify GA-related targets,on the basis of which a drug-chemical component-target-lesion network diagram was established.Topological analysis of the drug targets was carried out through the STRING database,leading to the detection of core genes.In addition,GO functional enrichment and KEGG information channel resolution were carried out for the core prescription and patient common targets to gain insight into the GA interaction mechanism.Results:1.The results of the analysis of drug frequency,efficacy,four qi and five flavors,and the attribution of meridians: After this study,we screened three hundred and two prescriptions,involving a total of two hundred and twenty-nine medicinal flavors,with a total frequency of use of five thousand two hundred and forty-six times.Among them,the drugs Cichorium sanctum,Che Qian Zi,Chuan Niu Knee,Qin Pi,Red Peony,Di Long,Dendrobium,Cuscuta,Angelica,and Poria were the most commonly used.In addition,we also analyzed the efficacy of the drugs and found that they mainly involved heat-clearing drugs,wind-dispersing drugs,water-permeating drugs and wind-damp dispelling drugs.The proportions of the four qi were cold,warm,flat,cool and hot,and the proportions of the five tastes were sweet,bitter,pungent,salty,sour and astringent,while the attributed meridians were liver,kidney,spleen,stomach,lung,heart,kidney,gall bladder,intestinal and small stomach meridians.2.Results of drug association rule analysis: Under the parameters of support number of 60 and confidence level of 0.6,74 high-frequency drug combinations with the frequency of use above 59 times and 100 drug combination association rules were obtained,and their common drug pairs were Psyllium and Cyperus rotundus;Psyllium and Szechuan hyssop;Psyllium and Szechuan hyssop;Psyllium,Cyperus rotundus and Szechuan hyssop,etc.3.Results of clustering analysis of high-frequency drugs: eight drug combinations were obtained,of which the first category was Pterostilbene,water hyacinth,red peony,raw earth and peony bark;the second category was white peony,rehmanniae,psyllium,mountain cichlidium,significata and qinpi;the third category was maidenhair,schisandra,wolfberry,puffed wheat and turtle shell;the fourth category was ginseng,astragalus,fried atractylodes,danshen and black sesame;the fifth category was gardenia,scutellaria,gentian and huanglian.The sixth category is Scutellaria baicalensis,Radix Bupleurum,Radix et Rhizoma Qingxian,Ziziphiopogonis,and Phellodendron;the seventh category is Radix et Rhiziphiopogonis,Phellodendron,Cuscuta,Dendrobium,and Atractylodes;the eighth category is Cistanches,Mulberry,Epimedium,Phellodendron,and Yang Qi Shi.According to the actual clinical use as well as the efficacy of the core prescription for the second category of drugs.4.Results of network pharmacology study: The results showed that the core prescriptions of GA treatment mainly consisted of eicosanoids,n-tridecane,24-methylenecyclic atunol,solasodanol and darutizil B,while the core genes included AKT1,EGFR,ERBB2,FGFR2 and GRB2,the functions of which could be confirmed by network pharmacology study.After enrichment analysis,we found that the core prescriptions have molecular functions such as regulating non-transmembrane protein tyrosine kinase activity,promoting peptide hormone responses,and regulating cell projection membranes.In addition,KEGG pathway analysis showed that signaling pathways such as neurotrophin signaling pathway,relaxin signaling pathway,FC&RI signaling pathway,and EGFR tyrosine kinase inhibitor resistance are important factors for the role of core prescription.Conclusion:1、Professor Tong Ying commonly used top 10 high frequency drugs for the treatment of gouty arthritis are Shan Cixi mushroom,Plantago ovata,Chuan Niu Knee,Qin Pi,Red peony,Di Long,Dendrobium,Cuscuta,Angelica,Poria,etc.2、Top 10 commonly drug pairs used by Prof.Tong Ying for the treatment of gouty arthritis are Psyllium,Mountain Cichlidium;Mountain Cichlidium,Chuan Niu Knee;Psyllium,Chuan Niu Knee;Psyllium,Mountain Cichlidium,Chuan Niu Knee,etc.3、Professor Tong Ying’s according to the top 10 drugs and drug pairs algorithm core prescription for gouty arthritis consists of Bai Shao,Shu Di Huang,Plantago ovata,Shan Cixi mushroom,Datura significata and Qin Pi.4、Through the network pharmacological approach,we found that the core prescription of GA treatment is to regulate transmembrane protein tyrosine kinase activity,promote peptide hormone response,and regulate multiple signaling pathways through various potential active ingredients,such as eicosanoids,n-thirty-two,24-methylenecyclic atunol,solasodanol,and darutizil B,acting on genes such as AKT1,EGFR,ERBB2,FGFR2,and GRB2.and modulate multiple signaling pathways for therapeutic purposes.Studies suggest that core prescriptions for GA may exert their mechanism of action by modulating the inflammatory response.
