Objective:To explore effect of Mongolian cupping and bloodletting combined with acupuncture therapy on NF-κB pathway in Rats with Cough Variant Asthma model.Methods:78 SD rats were divided randomly into six groups like the blank control group,model group,Mongolian cupping and bloodletting therapy group,Mongolian acupuncture therapy group,Mongolian acupuncture combined with cupping and bloodletting therapy group and western medicine group,there are 13 rats in each group.The blank group was given an equal amount of normal saline,the other five groups were given intraperitoneal injection by 1ml of 10%Ovalbumin(OVA)to cause the allergic reaction on the 1st and 8th day,and motivated by inhalation solution 1%OVA solution started at the 15th day,20 minutes/day,once a day,7 times in total,to establish the cough variant asthma rats model.To provide the treatment from the 29th day,the blank and model group gave the same method and amount of normal saline,the Mongolian cupping and bloodletting therapy group conducted the cupping and bloodletting therapy at its mother lung and child lung points,the Mongolian acupuncture therapy group conducted the acupuncture therapy at its mother lung and child lung points 15min/d,the Mongolian acupuncture combined with cupping and bloodletting therapy group conducted the acupuncture therapy combined with cupping and bloodletting therapy at its mother lung and child lung points,the western medicine group gave the budesonide suspension by atomization,treatment for a total of 9 days.Killing the rats within 24 hours after the last treatment,collecting the blood serum and taking the lung tissue samples.To inspect the shape of lung tissue by HE staining.The standard of blood serum eosinophil cationic protein(ECP)、transforming growth factor β1(TGF-β1)and interleukin-1β(IL-1β)checked by Elisa testing.The protein expression of NF-κBp65和 IκB in lung tissue tested by Western blot method.Results:1.The comparison of cough frequency show that:compared with the blank group,the model group rats’ cough frequency obviously increased(p<0.05);compared with the model group,the frequency of cough in the Mongolian cupping and bloodletting therapy,Mongolian acupuncture therapy,Mongolian acupuncture combined with cupping and bloodletting therapy and western medicine groups apparently reduced(p<0.05),there is no clearly diversity between the four treatment groups(P>0.05).2.HE stain testing outcome reveal that:compared with blank group,the model group’s gathering of inflammatory cells and the proliferation of airway smooth muscle obviously increased;compared with model group,All the Mongolian cupping and bloodletting therapy,Mongolian acupuncture therapy,Mongolian acupuncture combined with cupping and bloodletting therapy and western medicine groups can reduce the infiltration of eosinophils and airway inflammation,there is no clearly diversity between the four treatment groups.3.ELISA testing outcome reveal that:compared with the blank group,the standard of blood serum the protein of eosinophil cationic(ECP)、transforming growth factorβ1(TGF-β1)、NF-κBp65 and interleukin-1 β(IL-1β)on model group were obviously increased(P<0.05);compared with model group,the levels of serum eosinophil cationic protein(ECP)、transforming growth factor β1(TGF-β1)、NF-κBp65 and interleukin-1β(IL-1β)of the Mongolian cupping and bloodletting therapy,Mongolian acupuncture therapy,Mongolian acupuncture combined with cupping and bloodletting therapy and western medicine groups were reduced(p<0.05),there is no clearly diversity between the four treatment groups(P>0.05).4.Western Blot testing outcome reveal that:compared with blank group,the model group’s expression of NF-κBp65 and IκB protein was significantly decreased(P<0.05)in Lung;compared with model group,All the Mongolian cupping and bloodletting therapy,Mongolian acupuncture therapy,Mongolian acupuncture combined with cupping and bloodletting therapy and western medicine group’s expression of NF-κBp65 in Lung was obviously reduced(P<0.05),the Mongolian acupuncture therapy,Mongolian acupuncture combined with cupping and bloodletting therapy and western medicine group’s expression of IκB protein was significantly decreased(P<0.05)in Lung,the IκB protein expression of Mongolian cupping and bloodletting therapy was no clearly diversity(P>0.05).Conclusion:1.Mongolian acupuncture combined with cupping and bloodletting therapy can apparently improve the cough symptoms of CVA model rats,lighten the infection of inflammatory cells and reduce the level of ECP in serum,thus reduce the airway inflammation.2.Mongolian acupuncture combined with cupping and bloodletting therapy can constrict the activation of NF-κB signaling pathway,reduce the NF-κB and IκB protein expression in CVA model rats’ lung tissue;meanwhile,it can inhibit the NF-κB signaling pathway to release inflammatory factors like IL-1β and decreased the level of IL-1β,thus inhibit airway inflammation. |
