| ObjectiveGinger goat broth of angelica sinensis was established by zhang zhongjing of the eastern han dynasty as a classic medicine diet formula.This study used angelica ginger sheep broth as a positive control in the female rat model of spleen kidney deficiency syndrome,compared with jinwu broth.MethodsMedicinal angelica ginger and goat broth is cooked to the exact recipe described in the Chinese Prescription Dose Book.The broth is then filtered for food scraps,divided between the meat and broth and stored in a fridge at 4 ° C.Seventy-two healthy SD female rats with SPF clean were included in the study,with a mean weight(200 ± 20)g and no abnormal performance after one week of adaptive feeding.Randomly divided into normal group,spleen,kidney and yang deficiency model adenine and cathartic leaf aqueous decoction combined to cause spleen,kidney and yang deficiency model.After the successful molding,the positive group was fed Jinwu soup,the treatment group was given low,medium and high corresponding concentrations and proportions of angelica ginger goat broth soup and meat,and normal and model groups were fed with isometric saline and normal feed.The trial lasted 10 weeks,the molding phase 6 weeks and the drug intervention phase 4 weeks.The experimental indicators were as follows: 1.During the experiment,the general state of each group of rats was observed daily(including mental state,hair,feces,etc.).2.Weight and anus intake were measured weekly in each group.3.The rats were subjected to an absenteeism experiment every two weeks during the dosing comprehensively,and the possible mechanism of action was discussed.Results1.Animal model: animal model of spleen,kidney and yang deficiency was prepared by adenine and catharsis aqueous decoction.After 6 weeks of castration,rats showed lethargy,thinning or shedding of hair,palpable coldness at claw touch,no blood color,pallid tongue moss,soft stools,or purulent stools.There was a significant difference in body weight between the normal group and the model group after castration,with significantly lower body weight compared to the normal group(P <0.05),significantly lower anal temperature compared to the normal group after caudal motility compared to the normal group(P < 0.05).2.In normal group,the mental state,hair and fecal matter were normal.The rats in the model group showed lethargy,yellowing and thinning of hair and lack of luster.Compared with the model group,there was different improvement in all treatment groups,especially in the high-dose group of angelica ginger goat broth.3.Weight and anal moderate intake: in addition to the normal group,the anal moderate weight and intake of rats in the model group,the positive group and the angelica ginger sheep broth were all reduced in the low,medium and high doses.After administration of the drug,weight gain and anal temperature increased in all treatment groups.The anal temperature was significantly higher in the positive group than in the model group,with significant differences(P < 0.05).The anal temperature in the mid-dose group was significantly higher than in the high-dose group(P < 0.05).4.Absence experiment: Normal group rats moved quickly in the box and frequently passed through the central region.Compared to the normal group,the rats in the model group showed significantly reduced activity,longer stay in the corner region,longer total rest time,significant difference(P < 0.05),shorter total movement distance,shorter rapid movement time,and significant difference(P < 0.05).All treatment groups showed greater improvement in activity after administration of the drug compared to the model group,with lower total rest time in the positive group and lower,higher medium and higher doses of angelica broth,and higher central movement time in the rats with shorter and higher doses(P < 0.05).5.RGB values: tongue,claws and tail redness in normal rats.The tongue,claw,and tail were paler or paler in the model group,and the tongue,claw,and tail were redder in each treatment group than in the model group.The positive group had lower tongue,claw,and tail values of R,G,and B than in the model group(P < 0.5),while the high dose of G was significantly higher in the high dose group(P < 0.5),and the high dose of G in the high dose group(P < 0.05).6.Red blood cell(RBC)and hemoglobin(HGB)levels: RBC and HGB levels were lower in the serum of rats in the positive group than in the normal group,and RBC and HGB levels were higher in the serum of rats in the positive group than in the medium and high dose group of angelica ginger broth(P > 0.05).7.Urea nitrogen(BUN)and creatinine(CRE)levels were higher in the serum of rats in the positive group than in the normal group,and there was no significant difference in serum BUN levels between the positive and angelica ginger goat broth in the low,medium and high dose groups compared to the model group(P >0.05).However,the serum CRE content in the model group was higher than that in the normal group,with significant differences(P < 0.05).Compared to the model group,the serum CRE content in the positive group and the medium and high dose group of angelica ginger goat broth was decreased,and there was no significant difference between the groups(P > 0.05).8.Elisa assays: Serum levels of E2(estradiol)and c AMP(phosphoryldiesterase)were lower in the model group than in the normal group(P > 0.05).Serum levels of E2 and c AMP were increased in the positive group and in the medium and high doses of angelica goat broth compared to the model group(P > 0.05).9.Visceral wet weight: The spleen was heavier in the model group than in the normal group,and there was no significant difference(P > 0.05).The spleen wet weight decreased in the positive group and in the medium and high doses of angelica ginger broth compared to the model group,but there was no significant difference(P > 0.05).Compared to the normal group,rats in the positive group showed a significant difference in kidney wet weight(P < 0.05),and rats in the positive group showed a decrease in kidney wet weight compared to those in the medium and high doses of angelica ginger broth,with significant differences between the positive group and the high group(P < 0.05).ConclusionsThrough adenine and catharsis aqueous decoction,the rat model of spleen,kidney and yang deficiency syndrome can be successfully caused,angelica ginger yang broth can improve the general condition of spleen,kidney and yang deficiency syndrome rats,promote blood production,improve blood circulation,repair kidney injury,improve the overall situation of spleen and kidney yang deficiency syndrome rats. |
