Fluorescence method is a hotspot in modern measurement filed. It can distinguishclassify and thickness of substance by measuring special fluorescence spectrumcharacteristic of them, and has wide application foreground. Phytoplankton in the sea isthe most important primary producer and the primary energy converter in sea ecosystem.For alga is the most common phytoplankton, identifying them can estimate thecommunity structure and distribute status of ecosystem in the sea area and realize theinspecting and comprehensive father of sea. Base on the analysis and synthesis of alga status inspecting technology anddevelopment trend in the world, after analysis on representative fluorescence spectrum oftetraselmis chuii,chlorella vulgaris,dunaliella salina which belong to chlorophyta,haetoceros,nitzschia closterium which belong to bacillariophyta, dicrateria sp whichbelong to chrysohyta. This paper brings forward ratio fluorescence spectrum andfluorescence gross method based on algae pigment;two order differential coefficientspectrum and fuzzy clustering analysis method based on super micro structure difference;linear multi-element regress analysis method base on single class algae and mixed algae'sdifference coefficient statistics characteristic. And after identifying experiment on singleclass and mixed algae solution of six classes algae in laboratory, an ideal identify resultwas got. The primary research content as follow:(1) Based on primary theory of fluorescence measurement, algae fluorescencemeasurement mechanism was researched, this provide manner and experimentfoundation for identifying different class algae.(2) Experimental research has been done for absorb spectrum,excitation spectrumand fluorescence spectrum character of different class and strength of alga solution. Thebest excitation wavelength and fluorescence wavelength for alga identifying was decided.(3)The wavelet method for fluorescence emission and excitation spectrum noisereduce was researched, which can reduce spectrum signal distortion and useful signalattenuation, the spectrum signal and noise ratio and flatness was enhanced.(4)Algae fluorescence spectrum excitation by pigment characteristic light,two orderdifference coefficient of fluorescence spectrum excitation by 440nm light andfluorescence gross method for single class algae characteristic pick-up was researched.And brings out fuzzy cluster analysis method which use single algae fluorescencespectrum's two order difference coefficient grain point as characteristic point. At thesame time, single algae's cluster was analysis.(5)Based on single algae and mixed algae excitation fluorescence spectrum'sstatistic characteristic of two order difference coefficient, multi element linear regressmodel was researched which used algae fluorescence spectrum two order differencecoefficient peak point and grain point, algae component was analysis.(6)In the laboratory, experiments for identifying six class different algae werecarried through. Reasonable of characteristics pick-up and identify model is affirmed.The primary innovation of this paper is that combining fluorescence method withration spectrum analysis,fluorescence gross analysis,fuzzy cluster analysis and modeidentify, reduces the complex of identify method, provides a practical design schematicfor identify system;single algae's fluorescence excitation spectrum ration characteristicpick-up,difference coefficient characteristic pick-up,fluorescence gross characteristicpick-up,algae fluorescence excitation spectrum two order difference coefficient graincharacteristic pick-up and mixed algae two order difference coefficient fluorescenceexcitation spectrum characteristic method is researched;carries through multi elementlinear regress analysis based on linear splice model, mixed algae's fluorescenceexcitation spectrum satisfying linear summation was discovered. The experimentresearch further validates comparability of single class and mixed algae's two orderdifference coefficient fluorescence excitation spectrum and super micro structure.Applying all the method mentioned here for fluorescence alga inspecting system, afew different classes single or mixed algae component can be identified. This paper gavea novel technology for alga inspecting.
