Discussion Of The Influence Of Nano Pulverization On Structure Chemical And Pharmic Characters Of The Liuwei Dihuang

High Speed Cutting and Sheering pulverizer is a newly developed apparatus, which has an entirely different construction compared with agitating mills or ball mills. On the sub axis, there are a number of rings-shaped grinding media, which can move freely. With the rotation of the axis and the autorotation of the rings, powerful centrifugal and cutting force can be produced to pulverize the Traditional Chinese medicine (TCM) to sub-micron or nano particles within a short time. It would take profound influence on the pharmacy of TCM. In this paper, nano Liuwei Dihuang is prepared by the technology. The performances of nano medicine are studied systematacially with the guidance of traditional theory.Using Liuwei Dihuang as raw material, the appropriate process parameter of preparing nano particles was studied. The particles were characterized by ZetaPALS light scattering granulometric analyzer, optical microscope and SEM. Experimental results show that the average diameter of nano particle is 161.9nm and the particle diversion is equably. The great majority of the plant cell is broken after nano pulverization. The effective components could be released directly. Results indicates that the dissolution of 80 mesh particles at different time increase with time and the difference of the dissolution at different time is statistically remarkable, while the dissolution of nano particles behave no relation with time and the difference of dissolution at different time isn't remarkable. The accumulative dissolution of Liuwei Dihuang is improved by nano pulverization.IR and HPLC chromatograms of Liuwei Dihuang illuminate that the structural feature of nano particles'active ingredient has not been changed and the extraction percent of paeonol from Liuwei Dihuang is increased by 23.52% after nano pulverization. A new opinion is brought forward that HSCS pulverizer is a new method to extract active ingredients from plant medicine. No organic solvents are added in the whole process.The maximal concentration (C_max) of paeonol of nano particles in rats is 61.6ug/mL, which is higher than that of 80 mesh particles (32.7μg/mL).
