Study On The Clinical And Mechanism Of Huoxue Jiedu Decoction In Treating Sjogren 's Syndrome

Sjogren’s syndrome (SS) is a chronic inflammatory autoimmune disease,mainly involving the humanexocrine gland. Clinical in addition to dry mouth,dry eye symptoms and signs,the performance of multi system damage is the respiratory system,urinary system,nervous system,blood system,digestive system,endocrine system. Because of its etiology and pathogenesis is still not clear,so give treatment brings certain difficulty.Part 1 Literature reviewThe present research on various aspects of SSetiology, pathogenesis, diagnosis and treatment of traditional Chinese and Western medicine and the latest progress of summaryinduction.Part 2 Clinical researchObject:Through the observation of clinical curative effect of HuoXueJieDu Fang(HXJDF) on patients with primary’s Sjogren syndrome (pSS) and improved quality of life,to explore the mechanism of HXJDF in the treatment of pSS.for its further application in clinic lay the theoretical foundation.Methods:84 patients with pSS were randomly divided into treatment group and Control group,42 cases in each group.The treatment group was treated with HXJDF,and the control group was treated with total glucosides of paeony(TGP).The treatment period is for 8 weeks,at the same time to record the changes of TCM syndrome before and after treatment. Two groups of patients were detected physical examination and laboratory indexes,and assessment of the quality of life questionnaire score before and after treatment.Results:After treatment,the treatment group total effectiveness is 82.50%,and the control group effectiveness is of 74.36%,which there is no significant difference between the two group(P>0.05).The two groups after treatment can reduce the total and signal TCM symptoms score, which in improving dry eyes and dry eyes integral,the treatment group is better than the control group (P>0.05). The two groups after treatment can reduce CRP,immunoglobulin(IgG,IgM,IgA) and increase tears,saliva secretionthe and prolong the tear break-up time. The treatment group also can reduce ESR,whole blood low shear viscosity levels(P<0.05).The two groups after treatment can improve the SF-36 in part dimension score,and reduce the SAS score (P<0.05).Conclusion:HXJDF in the treatment of pSS has clear curative effect, not only can reduce the TCM symptoms integral,improve physical and laboratory examination indexes,and can also improve the quality of life.Part 3 Experimental studyObject:To investigate HXJDFHuoxue on NOD mice experimental study on TLR2,TLR4,CD14 signal transduction pathway.Methods:6 C57BL/6J mice were set as normal group, and 18 NOD/Ltj mice were randomly divided into model group,Chinese medicine group (HXJDF group) and control group(TGP group). After 8 weeks,mice were measured the salivary secretory volume,submandibular gland index,and the measurement of serum and submandibular gland of mice were respectively TLR2,TLR4,CD14 protein and mRNA.Results:Compared with the normal group,the increase of TLR2, TLR4,CD14 protein and mRNA expression of the remaining three groups of mice serum and submandibular gland(P<0.05). Compared with the model group,Chinese medicine group and control control group to reduce TLR2,TLR4,CD14 protein and mRNA expression of mice serum and submandibular gland(P<0.05).And there is no difference in the traditional Chinese medicine group and the control group(P>0.05).
