Objective: The prescription of Rekeping injection is commonly useddrugs in clinic. It made up of Centipeda minima and Bupleurum chinenseDC, the antipyretic and anti-inflammatory effects are remarkable. TCMhas clearly effect and its side effect is small,but the compositions of TCMare complicated. In recent year, security incidents have occurredfrequently and how to improve the security and effectiveness of traditionalChinese medicine injection (TCMI) has been the Research focus. Theresearches about Rekepin are insufficiency, both at home and abroad. Theeffective compositions of Rekepin are not clear. To determine the activecompositions which have antipyretic and anti-inflammatory effect, toimprove the security and effectiveness of Rekepin, The study was carriedout by analyzed the volatile of Centipeda minima and Bupleurum chinenseDC. The aim of the study is to adjust ratio of Centipeda minima andBupleurum chinense DC based on original prescription, to optimize itssafety and effectiveness.Method: First of all, extracted volatile from Bupleurum chinense DCby Steam distillation, separated VOBC by silica gel columnchromatography. Then, we made the VOBC injections and components separated injections by refer to quality standard of Bupleurum chinenseDC injection. We utilized the raised extent of rabbit temperature caused byendotoxin, observed the antipyretic effect of Bupleurum chinense DCvolatiles and its separated components, utilized the swelling extent of rattoes induced by egg white and the swelling extent of mice ears caused byxylol, we observed the anti-inflammatory effect of VOBC and itsseparated components; the abirritation of VOBC and its separatedcomponents was assessed by hot plate plan. Thirdly, analyzed thecomponents by GC-MS.Finally, adjusted the ratio of the two herbs basedon the data of research of Centipeda minima and determined the best ratioof the active components of Centipeda minima and Bupleurum chinenseDC volatiles by animal experiment.Result: The Bupleurum chinense DC volatiles was separated to5parts by silica gel column chromatography and marked asVOBC-(1-5).The VOBC and VOBC-2showed obvious inhibition onraised temperature of rabbit caused by endotoxin、 swelling rat toesinduced by egg white、swelling mice ears caused by xylol and pain causedby hot plate plan. The antipyretic effect of VOBC-2is stronger than theVOBC. However, the difference is not clear between the VOBC-2and theVOBC in anti-inflammatory effect and abirritation. The results showed theVOBC-2are the antipyretic、analgesia and anti-inflammatory effectivecomponents of VOBC. The best ratio of antipyretic is8:2(Centipedaminima: Bupleurum chinense DC).Conclusion: The main effective material basic of Rekeping injectionhas been found and the best ratio of extraction process and prescriptionwere determined by the experiment. The result of the experiment providesa certain basis for development of new Rekeping injection.
