Objective Graves' disease (GD) is an autoimmune thyroid disorder involving on antibody (TSAb) directed against the TSH receptor (TSHR) producing thyroid stimulation. Its symptoms and signs include sweating, nervousness, fatigue, insomnia, sensitivity to heat, hunger, less of weight, diarrhea, trembling hands, and bulging of the eyes. Often, there is a swelling in the neck. In order to improve and perfect the method of making Graves' disease, animal model, probe deeply curative mechanism of Tablet-FFJK, and screen new Chinese herbs of antithyroid, we have used new theories and methods of modern medicine to make experimental study of GD. MethdosThe model was reconstructed: The Wistar rats were mainlined O.lml, 0.2ml, 0.3ml, 0.4ml, 0.5ml Y.E bacteriurm water (103/ml)in trail in first, fifth, tenth, fifteenth and twentieth day. The symptom was observed since seventh day and the blood and thyroid gland were drawed in twenty-fifth day. The FTa> FT,? TTs and TT4 were measured.(2) The curative mechanism experiment The 300% Chinese herbalmedicine water was made, and mice was fed with it for 0.2ml/10g one month. The FT3^ FT4, TT3 and TT4 were measured.(3)In this experiment, we divided the Wistar rats into Normal group, model group, Traditional Chinese medicinal group, western medicinal group, Tablet-FFJK 1, Tablet-FFJK2, and DNM at random. These rats were cured one month. Their bleed and thyroid glands were drawed. The FT3, FT4> TTa and TT4 were measured by RIA, the expression of thyroid MAPK was observed by immunetochemistry dyeing, the thyroid NIS, c-fos, TGF- P i mRNA were observed in situ hybridization and the histological changes were observed by light and electronic microscopes. ResultsIn the model group, the serum levels of FTj, FT4, TTs and TT4 was higher than Normal group (P<0.001). The results of common pathology study showed that the majority of thyroid follicle epithelial cells appeared with cube shape and the minority low pole. The capillary among intertitial tissue dilated. There was sorb vacuole in some follicle.(2) The serum levels of FT^^ FT4, TTs and TT4 DNM group were obviously lower than the Normal group (P<0.01).(3) The study of symptom and signs showed that the rats of model group acted more frequently, drunk more water, and were mental disturbance in the nonage, and become thin, acted for shorter time and stool was watery after the model was formed. The quantum of water and feed was oberviously higher thanNormal group (P<0.001). their weight was lower than Normal group (P<0.001). The symptom and signs in the curative groups oberviously improved, the most significant was FFJK2.(D The RIA measure results showed the serum levels of FTs, FT,? TTs and TT4 in the model group obviously increased, and decreased in all curative groups.(5) The results by IHC showed that the expression of rat thyroid MAPK in the model group was High. The other groups were lower expression than it, the most significant among them was FFJK2.(6) The results in situ hybridization showed that the expression of NIS > c-fos mRNA were high, and TGF- P i was low. The results were changed by the curative groups, the most significant among them was FFJK-2. The study of common pathology showed that the follicles are small and lined with hyperplastic columnar epithelium and contain scant colloid that displays much marginal scalloping and vacuolization. The nuclei are vesicular and basally located and exhibit occasional mitoses, papillary projections of the hyperplastic epithelium extend into the lumina of the follicles. Vascularity is increased, and there is a varying infiltration by lymphocytes and plasma cells.The instance above improved in all curative groups, and the most effect among them was FFJK2.(8) The ultra structure of thyroid showed that the microvillus in thyroid follicle epithelial cells increased in the model rats, and there were plenty of Mitochondria, lysosome, Rough endoplasmic reticulum, The Euchromatin increased in the nucleus.
