Experimental Study On The Effects Of Ovary â…¡ Formula On Hypophysis, Adrenal Gland And Ovary Of RAT With Polycystic Ovarian

In this project, we made pathological model of rat and obverted the influence of Ovary II formula on hypophysis, adrenal gland and ovary of rat with polycystic ovarian with optic microscope, electronic microscope, radio-immunity immunological histoc hemis-try and In situ hybridization The results: (1)Ovary II formula had effect of reducing LH, LH/FSH, T and increasing FSH, E2 in blood serum of rat with polycystic ovarian, which indicated that in has regulating effect on H-P-O. (2) Ovary II formula has effect of regulating morphological change of hypophysis of rat with polycystic ovarian; inhibiting reticular to produce androgenic hormones, making cellular layer of theca of follicle thicker and ovary granular cell thinner, promoting cell apoptosis of theca folliculi, inhibiting production of androgenic hormones. (3)Ovary II formula can promote expression of Bcl-2, Bcl-2 mRNA of ovary granular cells of rats with policytic ovarian, reduce expression of Bax, Bax mRNA, Fas, Fas mRNA, which indicated that it increase Bcl-2 from gene transcription to translation, and also reduce Bax, Fas from transcription to translation, it has double regulating effects and inhibiting granular cells apoptosis. (4) Ovary II formula can reduce IGF-1R, IGF-1 mRNA of follicle cells of rats with pollicytic ovary, which showed that Ovary II formula can reduce IGF-1R through inhibiting IGF-1 mRNA expression, IGF-1 transcription and production of androgenic hormones.This experiment proved that Ovary II formula has effect of inhibiting LH and androgentic honmones, anti-androgentic and IGF-1, inhibiting ovary granular cell apoptosis and promoting ovulation, which indicated that it has many regulating functions.Beside regulating H-P-O and H-P-A, Ovary II formula also has local regulating effect, etc. which showed that Ovary II formula took effect through microcosmic regulations of many sites, the character of diffusion and diversity with microscopical oneness, balance is advantages of Ovary II formula in treating PCOS.This study provide theoretical evidence to treat PCOS and laid foundations for new drug study.
