Back ground: Recent studies showed that icariin has the biological effect on stimulating osteoblast proliferation and differentiation. Object: To explore the effect of icarrin on stimulating Smad4 mRNA level in MC3T3-E1 cells.Methods: MC3T3-E1 cells were divided into four groups by different icariin stimulating concentration of 0ng/ml, 10ng/ml, 20ng/ml, 40ng/ml. MTT method and population doubling time were applied to observe the cell proliferation. To evaluate cell differentiation, the cell ALP level was determined by atomic absorptiometry methods, and the type I collagen was observed by immunohistochemistry. FinallyRT-PCR was engaged to find the Smad4 level in the MC3T3-E1 cell under the different concentration of icariin stimulation. Statistic analysis was carried out by SPSS software.Results: (1) under the invert phase contrast microscopy, MC3T3-E1 cell oligodendro-shaped with prosperous cell organs, with chromatin losely arranged in the nucleus were observed. (2) cell growth curve in lOng/ml group was higher than other 3 groups and the 20ng/ml group was higher than Ong/mK 40ng/ml groups. The population doubling time in the lOng/mlgroup was shorter than all other three group, 20ng/ml group was shorter than Ong/ml, 40ng/ml groups, the the differences above were all significance (P<0.01). (3) there were no any significance in the level of ALP among the 4 groups during the first day of culture.
