Objective:(1)Tooptimize the extraction process of Artemisia Volatile Oil.(2)To analyze the constituents of volatile oil in Artemisia Volatile Oil from different area and to compared with each other.(3)To raise the solubilizing capacity of Artemisia Volatile Oil by selecting the preparation procedure.(4)To study the anti-inflammatory and anti-allergic effects of Artemisia Volatile Oil.(5)To establish mouse ear edema model,permeability of mice abdomen capillaries models and delayed type hypersensitivity in mice,investigating the anti-inflammatory and anti-allergic effects.(6)To establish allergic rhinitis(AR)rats models,investigate the therapeutic effects of Artemisia Volatile Oil with allergic rhinitis models.Method:(1)The volatile oils of Artemisia in three different area were extracted by water vapor distillation and studied by means of the GC-MS,the components were quantitatively determined with area normalization method.(2)Using single factor design to determine the content of oil yield as the index of evaluation.(3)Using single factor design to determine the solubilization capacity of Artemisia Volatile Oil as the index of evaluation.(4)The effects of Artemisia Volatile Oil of analgesia were observed by mice inflammation models caused by dimethyl benzene and mice models with reinforce permeability of abdominal cavity capillary caused by acetic acid.(5)The mouse model of delayed type hypersensitivity was established by sensitizing the skin of Kunming mice with 2,4-dinitrochlorobenzen(2,4-DNCB).(6)Using ovalbumin(OVA)systemic sensitization and nasal antigen challenge,establish the model of allergic rhinitis in rats.Study on effects of volatile oil of Artemisia samples of rat serum IgE,sIgE,IL-12 and nasal mucosa structure,mast cells,eosinophil,basophil and other conditions by using enzyme-linked immunosorbent assay(ELISA)method for the determination of serum immunoglobulin E rats(IgE),specific IgE(s IgE)and interleukin-4(IL-4)content,and histopathological examination of nasal mucosa.Results:(1)The best extraction process is the ratio of material to liquid 1:8,the crude powder of the raw material is 5h,the reflux is extracted 5h.(2)The main compounds and their contents were different in three different area.(3)Poloxamer188 as surfactant,and the ratio was 56:1,and the rest of the prescriptions of the mixed materials can be made stable appearance in Artemisia volatile oil solution.(4)The volatile oil samples of Artemisia of each dose group can inhibit xylene are significantly different(P<0.05)induced mouse ear swelling.(5)The volatile oil of Artemisia samples in high dose group can significantly reduce the capillary permeability in mice,which are significantly different(P<0.05).(6)The volatile oil samples of Artemisia in each dose group can improve nasal symptoms and change AR animal model of nasal mucosa,reduce serum IgE,s IgE and IL-4.Conclusion:(1)The results can provide theoretical basis.(2)In theory,preliminarily explant in the same picking time,chemical constituents and contents of essential oils were different types in different area.(3)According to the design parameters determined by the study,the prepared samples were homogeneous and stable.(4)The volatile oil samples of Artemisia has obvious anti-inflammatory and anti-allergic effects.(5)The essential oil of Artemisia samples could inhibit AR synthesis of IL-4 rats,reduce serum IgE and s IgE levels of rats with allergic rhinitis,and improve the pathological changes of nasal mucosa.(6)Artemisia Volatile Oil has certain therapeutic effect on allergic rhinitis in rats.
