Background and objectionNow the pathophysiological mechanisms of restenosis have been considered to including both migration and proliferation of SMC, and elaboration by these cells of abundant extracellular matrix leading to progressing vascular remodeling. Tongxinluo is a compound Chinese herb which contains eight kinds of ingredients; Renshen, Shuizhi, Quanxie, Tubiechong, Wugong, Chantui, Ch-ishao and bingpian. The compound of these ingredients has a series of functions, including antiplatelet, anticoagulation, and blood vessel dilatation, maybe has lipid - modulating effect. Previous studies suggested that Tongxinluo has the inhibitory effects on the formation of atherosclerosis and the proliferation of vascular smooth muscles after injuried. Our study was submitted in rabbit of atherosclerosis narrow vessel model, to observe the inhibitory effects of Tongxinluo on intimal proliferation and vsacular remodeling and its possible molecubiological mechanisms after angioplasty in rabbit.Materials and Methods1. Animal model ;60 male white rabbits (2.5 -3.5Kg; 6 -8months) , All the animals were divided into 3 groups: control group ( normal diet plus intimal injury) , high -lipid group (high cholesterol diet plus intimal injury) , Tongxinluo group (Tongxinluo 0. 5mg/Kg/d, 4 weeks, also high cholesterol diet plus intimal injury). After hypercholesterol diet (1. 5g/Kg/d) for4 weeks, then the abdominal aortic endothelium were peeled off by balloon. The high - lipid group and Tongxinluo group continued to have hypercholesterol diet for another 4weeks and followed by abdominal aortic angiography. The narrow parts of the vessel underwent angioplasty until being satisfied dilated with no stenosis, contrasting with adjacent unnarrowing parts. After 4 weeks all rabbits were killed, and vessel samples were excised, respectively for the detection of histomorphom-etry, immunohistochemical, and molecubiological detection.2. Determination of blood sample; the blood sample were collected form the peripheral vessel 4 times ( before high - lipid diet, 4 week before injuried , 8 week before PTC A, 12 week before killed) . The concentration of TC( total cholesterol) , TG (triglycerin) , HDL - C ( high density lipidprotein cholesterol) , LDL - C (low density lipidprotein cholesterol) , ET ( endotheline ) , NO ( nitrix oxide) , SOD(superoxide) , TPA(tissue plasminogen activator) ,PAI(plasmino-gen activator inhibitor) were detected.3. Pathological Analysis;The angioplastic part of the femoral artery was cut off and divided into 3 parts. One part of the vessel specimen was dehydrated, embedded and section-cut for morphologic studies and immunohistochemical a-nalysis. Morphological studies include HE staining, VG staining and verheroff staining, The lumen area, intimal area, internal elastic lamella area (IEL) and external elastic lamella area (EEL) were detected by MetaMorph Image System after staining.4. Immunohistochemical analysis include PCNA ( proliferating cell nuclear antigen ) , MMP2 ( matrix metalloproteinase2 ) , MMP9 ( matrix metalloproteinase9) , TIMP1 ( tissue inhibitor of metalloproteinsael ) , TIMP2 (tissue inhibitor of metalloproteinsael) , ERK1 ( extracellular signal regulator ki-nasel) and ERK2 ( extracellular signal regulator kinase2 ) , by the monoclonal antibody SP method. The positive cells were stained brown, we detected the light density and area of positive cells by MetaMorph Image System after staining.5. RT - PCR (reverse transcription polymerase chain reaction) was done to detect the mRNA level of TIMP1, TIMP2, MMP2, MMP9. Relative level of MMP/TIMP = MMP/TIMP mRNA belt density/GAPDH mRNA belt density 6. The expressing of ERK1 and ERK2 was detected by Western Blot. The relative light density of them were analysis by fluorescent light scanning.Statistic AnalysisAll the data were expressed as mean valus ?standard deviation.
