Objective To identify and characterize mesenchymal stem cells from synovial membrane in vitro. Methods Synovial Mesenchymal Stem Cells(SMSCs) were obtained by limited dilution method and expanded in 25 milliliter flasks. The expression of vimentin, keratin were detected by immunohistochemical staining, respectively, while the expression of CD8, CD34, CD44, VCAM-1 and ICAM-1 determined by flow cytometry. Results Pure mesenchymal stem cells were of spindle shape, uniform in size, which were intensive positive in vimentin, but negative in keratin. The expression of CD44, VCAM-1 and ICAM-1 were also verified by FCM. Conclution Mesenchymal stem cells can be purified from synovial membrane in TMJ.
