Clinical study: Objective: To assess the efficacy of Traditional Chinese Formula NaoKangNing(NKN) in Patients with Parkinson's disease. Methods: 60 patients with PD were followed up in a single-blind, placebo-controlled, with randomized study for 12 weeks. All patients were diagnosed accroding to Western medicine diagnosis standard and Traditional Chinese medicine diagnosis standard. The patients were randomized divided into two grades, treating group and controlled group. Patients' characters, history, initial symptoms, UPDRS had no significant difference at the beganning. Patients in treating group were taken NKN capsule and taken placebo capsule in controlled group at the basement treatment. UPDRS score, H&Y classification, usage of madopar, on-off phenomenon, non-moter symptoms, quality of life were used to evaluate efficacy of the two groups in treatment of PD. Results: UPDRS score was significantly lower in NKN group than the begining (p<0.05). The H&Y classification of the treating group had changed. The usage of madopar decreased from 425. 5 + 296. 2 mg to 375. 5 + 225. 6 mg, and had significant difference than that of controlled group (P<0.01) . The effect of TCM symptoms in treating group was 76.67%, and had dignificant difference than 50.0% in controlled group(P < 0.05). The TCM symptoms of hypokinesia, muscle rigidity, bradykinesia, nape rigor, diziness, insomnia and dreamful sleep, sore waist, skelalgia and constipation in treating group has improved than that in controlled group. Compared with placebo group, the "off' time of patients in NKN group reduced 27% averagely; the "on" time prolonged 19% averagely. The quality of life had improved significantly in NKN group than that in plaedbo group. Conclusion: NKN capsule was effective in the treatment of PD espetially in UPDRS score, depression of the usage of Madopar, TCM symptoms,non-motor symptoms and quality of life.Experiment study: Objective: To study the mechanism of NKN capsule in treating PD. Methods: The model of Parkinson's disease was induced by intraperitoneal injection of MPTP into C57BL/6J mouse. The contents of dopamine (DA), malondialdehyde (MDA), nitric oxide synthase (NOS), and reduced glutathione hormone (GSH) in nigra striatum of model mouse were determined. Results: NKN capsule prevented the decrease of the contents of striatal DA, which indicated that NKN had neuroprotection on impaired PD mouse induced by MPTP. NKN significantly prevented the increase of the contents of MDA and NOS, while decreased the contents of GSH in SN induced by MPTP. These results suggested that NKN could scavenge the free radicals and has anti-oxidative effect on PD mouse induced by MPTP. Conclusion: Our results suggested that NKN, by the indirect modulation in the striatal DA and nigrostriatal anti-oxidative activity, prevents the neurotoxicity of MPTP.
