Methods:The inhibiting effect of Elemene on the HEp-2 cells and AMC-HN-8 cells wereevaluated by MTT assay. The apoptosis and necrosis cells were separated by FCM.The activity of Caspase-3 was assessed by colorimetric assay. The HEp-2 cells afterElemene treatment were analyzed utilizing Westernblot and RT-PCR. Results: HEp-2cells and AMC-HN-8 cells were significantly inhibited by Elemene, and the apoptosiscells increased with the increase of drug concentration and the elapse of time. Theactivity of Caspase-3 was enhanced after Elemene treatment. The expression of eIF4E,eIF4G, bFGF and VEGF protein were significantly inhibited by Elemene; and themRNA expression of bFGF and VEGF were inhibited either. Conclusions: Elemenecan effectively inhibit the growth of HEp-2 cells and AMC-HN-8 cells, and result inthe alteration of activity of Caspase-3. There were significant correlations between thedecreased expression of protein eIF4E, eIF4G, bFGF and VEGF. The mechanism ofeIF4E and eIF4G decrease the expression of bFGF and VEGF is post-transcriptional.
