Objective:By means of detecting the different expression level of AQP1(aquaporin1),VEGF (Vascular Endothelial Growth Factor) and MVD(microvessel density) in 60 cases of laryngeal squamous cell carcinoma(laryngeal carcinoma group),20 cases of para-cancer tissues(para-cancer group) and 17 cases of polyps of vocal cord (polyps of vocal cord group) as well as the dependablity during this three factors, to find out the clinical significance about AQP1 in the development of laryngeal carcinoma and the relationship with the angiogenesis of tumor ,to find out the possibility of AQP1 as the marker of laryngeal squamous cell carcinoma(LSCC) in order to early diagnosis,to judge the dependablity with the clinical classification,staging and the degree of differentiation,to investigate whether there is a significant difference in statistics between lymphnode metastasis group and non-lymphnode metastasis group in order to institude therapeutic regimen,make a decision of surgery ways and whether supplying radiotherapy,chemotherapy and other adjunctive therapy after operation. That is to provide scientific foundation for studying the pathogenesy of LSCC and directing the diagnosis,therapy and prognosis of LSCC.Methods:The immunohistochemical SP method was used to detect the expressionsof AQP1 and VEGF in tissue of each group, microvessel density(MVD) was also determined in endothelial as CD34.The patients were classified by International Union Against Cancer (UICC 1997) LSCC TNM Staging system .The data were analyzed by ANOVA, chi square test, rank test, and Spearman correlation. SPSS 13 system was used to perform the statistical analysis.Results:The expression of AQP1,VEGF and CD34 in the laryngeal carcinomagroup,para-cancer group and polyps of vocal cord group appeared to be different in degree , there was significant difference during the three groups(P<0.05), there was statistical significance between the laryngeal carcinoma group and para-cancer group, between the para-cancer group and the polyp of vocal cord group(P<0.01), but there was not statistical significance between the laryngeal carcinoma group and the polyp of vocal cord group for AQP1 expression,it told us there was close relationship between AQP1 and polyp of vocal cord oedema; The expression of AQP1 might be related to the clinical staging(P<0.01), lymph node metastasis (P<0.01), the degree of differentiation and sexuality (P<0.05), but no related to the clinical classification and age(P>0.05); The expression of VEGF might be related to the clinical staging(P< 0.01), lymph node metastasis (P<0.01) and sexuality (P<0.05),but no related to the clinical classification, age and the degree of differentiation(P>0.05);The expression of CD34 might be related to the clinical staging(P<0.01), the clinical classification (P<0.01) and the lymph node metastasis(P<0.05), but no related to sexuality, age and the degree of differentiation(P>0.05); There were positive correlation between the expression of AQP1 and VEGF(P<0.01)in the laryngeal carcinoma tissues ,the same to AQP1 and CD34, CD34 and VEGF.Conclusions:The high expression of AQP1,VEGF and CD34 in the LSCC and theclose relationship with TNM clinical staging, the clinical classification,the degree of differentiation and lymph node metastasis implicated that AQP1 may increase the vascular permeability of tumor, facilitate water fast transport within tumor cells,induce neovascularization and then promote tumor growth and metastatic dissemination. So the up-regulate of AQP1 suggest that AQP1 may play impotant roles in the laryngeal carcinogenesis and progression.In a word, AQP1 played an important role in the gro-wth,metastasis and angiogenesis of LSCC;The further investigation of AQP1 will benefit studying the pathogenesy of LSCC and directing the diagnosis,therapy and judging the prognosis.
