It was reported that hypertension had been 11.4 percent of all disease in our country in the first years of 90th in last century. Insulin resistance and /or hyperinsulinemia were/was "common soil" which were/was composed of hypertension, dyslipidemia, diabetes mellitus, cardiovascular disease and other associated metabolic diseases. So it has become a hotspot to investigate the relationship between insulin resistance and /or hyperinsulinemia and hypertension, to look for drugs that can decrease blood pressure and improve insuline resistance in preventing hypertension and some metabolic syndromes. Chinese traditional medicine has not been proved to play positive effects in preventing hypertension and the metabolic syndromes. At present, antihypertensive druggery have not been proved to take effective actions in preventing hypertension and the metabolic syndromes, let alone concomitance dyslipidemia and glucose intolerance. So we got a kind of mixture called "Tangzhixiao" by mixing extracts from modern medicine and Chinese traditional medicine. Tangzhixiao can reduce high triglyceride and cholesterol levels, improve IR and glucose intolerance and decrease blood pressure. Distinct toxicity and side effects had not been found in clinical test. In the study the molecular mechanism of insulin resistance and Tangzhixiao was elucidated by Radical Immunology assay and gene chip technology. We intended to find a new way to improve hypertension and contribute to open a new international market for Chinese traditional medicine.Objective Rat models of insulin resisting hypertension had been established and Tangzhixiao had been used in the rats. Then those factors relating to insulin resistance such as blood pressure, insulin, leptin, serum lipid have been measured and analyzed. The gene related to insulin resistance were separated by using gene chip and the effects of Tangzhixiao on relative genes of insulin resistance were elucidated, which was used to discussed mechanism of insulin resistance, Tangzhixiao preventing sensitivity of insulin from decreasing, increasing blood pressure and relative mechanism.Methods 4-5-week-old male Spraque-Darley rats were fed on high fructose diet to establish insulin resistant hypertensive rat model and then were randomly divided into five groups. The rats were given high fructose diet or high fructose diet containing three doses of Tangzhixiao and Rosiglitazone for 4 weeks. At the end of 4th week, the rats were sacrificed and their blood samples were taken. The concentration of serum glucose, insulin, leptin, lipids and erythrocyte calcium and magnesium were determined. The HOMA model was used to evaluate insulin resistance (IR). The relationships between parameters were analyzed. Soleus muscle from five rats of middle doses of Tangzhixiao ,Rosiglitazone and high fructose group were obtained. Total RNA was isolated and purified ,The chips were scanned with a Genepix Personal 4100A at two wavelengths to detect emission from both Cy3 and Cy5. The acquired images were analyzed using Genepix pro 4.1 software. Insulin resistance-associated genes were indentified and effect of Chinese traditional medicine Tang-Zhi-Xiao on insulin resistance was found .Results 1. SBP, insulin level and HOMA-IR in rats fed on high fructose were significant higher than those in normal control group, following hyperleptinemia and dislipidemia(p<0.05 or p<0.01).2. In rats fed on high fructose, HOMA-IR was strongly positively related to SBP, leptin level, triglyceride (TG) and erythrocyte calcium while negatively to high-density lipoprotein-cholesterol (HDL-C) and erythrocyte magnesium(p<0.05 or p<0.01).3. In rats fed on high fructose, leptin level was evidently related positively to SBP, insulin level, HOMA-IR and triglyceride (TG) (p<0.05 or p<0.01), but not correlated to serum glucose, total cholesterol (TC), HDL-C and erythrocyte calcium and magnesium(all p>0.05 ). 4.
