[Objective] Intrahepatic cholestasis of pregnancy(ICP) is a disease that occurs during the second half of pregnancy and does great harm to the fetus.The etiology and the pathogenesis are unknown. The studies have suggested that the estrogen may contribute to the pathogensis of ICP. During pregnancy, estrogens are produced and secreted by fetal-placental unit, otherwise placenta is as a target for estrogens. Our present study was designed to investigate the expression and the significance of ER α, ER β and PR in pregnant women with ICP. [Method] The kind of ER β mRNA on normal human placenta was investigated by reverse transcription-ploymerase chain reaction (RT-PCR) and the immunoreactivity localization of ER α and ER β on normal term gestation placenta was examined by immunhistochemistry (SP-assay);The expressions of ER α, ER β and PR were investigated by these methods. [Results] (1) There was mRNA expression of ER α and ER β on human term placenta, and the kind of ER β5 was expressed ; (2 )The ER α immunoreactivity on human term placenta wasin villous cytotrophoblastic nucleus, and that of ER P was in syncytitrophoblastic cytoplasm ; (3) The protein expression and the mRNA expression of ER P on placenta villi of pregnant women with ICP were significantly higher than those in normal pregnancy (P< 0.05),which the protein and the mRNA expression of ER a and PR were negative significance comparatively(P>0.05). [Conclusions] ER 3 5 was the kind of ER P on placenta, and it was expressed in cytoplasm of syncytitrophoblast and connecated with the production of estrogens probably ; the protein expression of ER a, ERP and PR on placenta were correlation with the mRNA expression of ER a ^ ER £ and PR ,and there was no expression block of ER a ?
