The Experimental Study On Xiaotan Sanjie Recipe Suppressing The Angiogenesis Of Gastric Carcinoma By Regulating The Expression Of Inducible Nitric Oxide Synthase

Objective To investigate the anti-angiogenesis effect of xiaotan sanjie recipe on the orthotopic transplanted gastric carcinoma of nude mouse bearing human SGC-7901 cell by regulating the expression of inducible nitric oxide synthase. Methods All the mice were divided into isotonic saline group, 5-Fu group, herb group and combination group randomly. Then the orthotopic transplanted gastric carcinoma model in nude mouse bearing human SGC-7901 cell was construced. The administrations began from the 2nd week after the model was constructed. The administrations were as follows: the mice in isotonic saline group were given isotonic saline 0.2ml ig. once a day; the mice in 5-Fu group were given 5-Fu dilution (60mg.kg^-1.w^-1) 0.2ml ip. once a week; the mice in herb group were given xiaotan sanjie recipe 0.2ml ig. once a day; the mice in combination group were given 5-Fu dilution (60mg.kg^-1.w^-1) 0.2ml ip. once a week and xiaotan sanjie recipe 0.2ml ig. once a day. The course of treatment was 4 weeks. When the experiment was terminated on the 11^th week all the mice were detected by ultrasound method to detect the blood supply. Then three-dimensional reconstruction was made to calculate the vessel volume and CD34 was stained to calculate the microvessel density to quantitative analysis the angiogenesis of the tumor. Immunohistochemistry and RT-PCR were applied to detect th expression of iNOS protein and mRNA. Results Xiaotan sanjie recipe can significantly inhibit the growth of orthotopic transplanted gastric carcinoma of nude mouse bearing human SGC-7901 cell. Xiaotan sanjie recipe could reduce the blood supplies of the tumors and reduce the calculation of MVD and downregulate the expression of iNOS protein and mRNA which suppress the angiogenesis of gastric carcinoma. Conclusion It maybe one of the possible anti-angiogenesis mechanisms of xiaotan sanjie recipe that it can downregulate the expression of iNOS protein and mRNA.
