| Objective To discover the restraining mechanism of Xiaotan Sanjie Recipe by way of observing the effects on the metastasis of gastric carcinoma and related adhesion molecules on orthotopic transplanted model. Methods 60 mice had been divided into control group, 5-Fu group, Xiaotan group, Sanjie group, Xiaotan Sanjie group and sham surgery group randomly before the orthotopic implanted model bearing gastric carcinoma cell line MKN-45 was built. The administrations began from the 3rd day after the surgery, which were as follows: the mice in control group were given isotonic saline 0.2ml ig. once a day; in 5-Fu group given 5-Fu dilution(60mg.kg-1.w-1) 0.2ml ip. once a week×3w; in 3 herbal groups given Xiaotan recipe, Sanjie recipe, and Xiaotan-Sanjie recipe 0.2ml ig. once a day respectively. The experiment terminated on the 6th week. The mice were sacrificed after blood sampling collection. The tumors and livers were weighted and the metastasis in liver was measured. E-cad, CD44v6, GMP-140, ICAM-1 and VCAM-1 were stained by immunohistochemistry. RT-PCR and Real-time PCR were applied to detect the expression of those molecules mRNA that encoded protein. Results Xiaotan Sanjie recipe can inhibit the growth of orthotopic implanted tumor and the metastasis in nude mice model bearing human MKN-45 cell line significantly. Xiaotan Sanjie recipe might reduce the expression of CD44v6, GMP-140 mRNA and encoding protein, which affect the metastasis of gastric carcinoma. Conclusion It might be one of the anti-metastasis mechanisms of Xiaotan-Sanjie recipe that it can down-regulate the expressions of CD44v6, GMP-140 mRNAs and proteins. The compounds in recipe might co-operate in cure.
