| According to the Traditional Chinese Medicine,improper diet,affection,work and rest result in organs function imbalance,air runless smoothly and fluid metabolism disorder.Among these etiological factors, "phlegm" is not only the material basis of produce carcinoma, but also a pathogenesis of carcinoma progressing and transferring. So it's an important way using the method of Xiaotan Sanjie to therapy carcinoma. We creat Xiaotan Sanjie Recipe according to the theory above described, and it indicated good effect on gastric cancer by clinical and experimental study.Metastasis is one of the most important characteristic of malignancy. Study on the molecular mechanism of tumor metastasis indicated that it is a complicated and multistepped process in which tumor cells and lodging cells make interaction. This process is under the charge of many special genes, which are activated or restrained and are interactional, which influence the biological characteristics of tumor cells eventually. We study the effect of Xiaotan Sanjie Recipe on expression of oncogene of gastric carcinoma such as ras and CerbB2 to further investigate the mechanism that Xiaotan Sanjie Recipe suppresses invasion and metastasis of gastric carcinoma.First all we establish the metastatic gastric carcinoma model of nude mice by orthotopic implantation of histologically intact human tumor tissue into gastric wall and observe its biological characteristics. Then we study the effect of treatment by Xiaotan Sanjie Recipe injected, 5-FU injected, normal saline injected. By Envision immunohistochemical method and semiquantitative reverse transcription polymerase chain reation, the expression of P21ras, P185, ras and CerbB2 in gastric carcinoma were determined.The main results are as follows:1.Gastric carcinoma orthotopic implantation model was established successfully. The study indicated its biological characteristics were similar to that of clinical gastric cancer andits metastatic potential was great. So it provides an ideal anmimal model for the study on human gastric cancer growth, infiltration and metastasis mechanism and antimetastasis therapy.2.The weight and the solidity of orthotopic tumor in Xiaotan Sanjie recipe group were significantly lower, comparing with those in control group. The inhibitory rate of tumor was 72.0%. The metastatic rate of distant lymph node and organs in Xiaotan Sanjie recipe group was significantly lower, too. These results suggest that Xiaotan Sanjie recipe could suppress the proliferation, infiltration and metastasis of gastric cancer cells.3.The expression positive rate of P21ras, P185, ras and CerbB2 in Xiaotan Sanjie recipe group were obviously lower than those in control group (P<0.05) by Envision immunohistochemical method and semiquantitative RT-PCR method. This is a hint that Xiaotan Sanjie recipe was likely to suppress the proliferation, infiltration and metastasis of gastric cancer cells by reducing the expression of oncogene CerbB2 and ras.
