Jin Long She Oral Liquor which is made from Xiaotan Sanjie Recipe has produced a good curative effect on clinic.Tumor proliferation and metastasis depend on angiogenesis. Prophase vitro experiment investigation in our department showed that Xiaotan Sanjie Recipe had inhibited expression of VEGF in gastric carcinoma. Based on the previous clinical and experimental research, we study the effect of Xiaotan Sanjie Recipe on expression of angiogenesis of gastric carcinoma such as VEGF and its receptor KDR To further investigate mechanism that Xiaotan Sanjie Recipe inhibits invasion and metastasis of gastric carcinoma.metastatic model simulating human gastric cancer was established by orthotopic implantation of histologically intact human tumor tissue into gastric wall of 15 nude mice. Total body condition and abdominal sign observed, nude were put to death when they were close to exhaustion.Tthoracie and abdominal viscera, lymph node were made H-E Staining to evaluate metastasis mode! successful or unsuccessful. And chromosome detect of the local and metastatical tumor were made to evaluate source of tumor. Metastasis model was established. Nude mice were abdominal cavity injected Xiaotan Sanjie Recipe.Then the local tumor weight, volum, inhibitary rates, the presence of metastasis and ascitic fluid formation were evaluated, we established an angiogenesis model of chick chorioSlantoic membrane to observe angiogenesis by Freeze-fracture electron microscope. By S-P immunohistochemical method and semi-quantitative reverse transcription polymerase chain reaction, the expression of VEGF and KDR in gastric cancer were determined. The main results are as follows:1.Gastric carcinoma metastasis model was made successful. It could reflect metastasis process of clinical stomach cancer, and offers an ideal animal model for human gastric carcinoma growth, infiltration and metastasis mechanism and antimetastasis therapy.2.Compared with the weight of orthotopic tumor in control group, all these in Chinese medicine group were significantly inhibited (p<0.05). The inhibitory rate of tumor was 72.00%. In addition, the incidences of liver metastasis were also significantly decreased in the Chinesemedicine group.All this produces were certificated that Xiaotan Sanjie Recipe could inhibit proliferation, infiltration and metastasis of gastric carcinomatous cell.3.In CAM experiment, the result of freeze-fracture electron microscope manifested that vessel in Chinese medicine group was obviously few than that of in control group.4.Expression of VEGF and KDR in Chinese medicine group were significantly lower than that of the control group by immunohistochetnical and RT-PCR method. It inhibited tumor angiogenesis. So tumor cell nutrien accommodation and its entrance to circle passage were blocked, thereby infiltration and metastasis of gastric carcinomatous cell were inhibited.
