In recent 20 years, the incidence of endometrial carcinoma (EC) has been increasing all over the world. EC has been one of the most common gynecologic malignancies. Therefore, EC will become one of the most important fields in tumor research. However, it has been recognized that intrinsic resistant to chemotherapy play an important role in the failure of chemotherapy in the patient with advanced recurrent or metastatic EC. So we try to research a new therapy route.Alterations in DNA methylation patterns accompany carcinogenesis and tumor development. Research for aberrant DNA methylation of tumor suppressor genes, DNA-methyltransferases and demethylation treatment are also hotspots. To analyze the role of P16 as a tumor suppressor gene in the genesis of EC and correlate lose of P16 mRNA expression to epigenetic mechanisms, we have performed a comprehensive study of P16 methylation status and P16 mRNA expressions in a series of 8 normal endometrial samples, 6 simple and complex hyperplasia, 6 atypical hyperplasia (AH) and 38 EC. Furthermore, we observed the effects of 5-aza-CdR on tumor growth inhibition, P16 exon 1 methylation, P16 mRNA expressions, Bax mRNA expression and cell cycle in the nude mice xenografted model SL-I of human EC.The mythylation status of the 5' CPG island of the P16 gene was evaluated...
