Glioma is a kind of malignant tumor that affects human health seriously with the characteristics of easy recurrence and difficulty in being eradicated. Current treatment of malignant glioma,consisting of gross surgical resection followed by radiotherapy and/or chemotherapy, has failed to substantially change the grim prognosis of patients.This has led to the exploration of other treatment modalities including biotherapy.Most of biotherapy methods are the study of tumor immune gene therapy on the principle of improving host immune response.Current immune gene therapy concentrates mostly on cytokine gene therapy. Many kinds of cytokines such as IL-2, TNF, IFN-γ have some antitumor effect in the past research. IL-4 is a molecule with mutiple biological activities acting on different cell types.To explore an experimental method of IL-4 gene therapy to glioma based on the principle of enhancing host immune response,and to understand the infiltrative growth features of glioma ,the growth characters of tumor cells transfected with foreign gene in vitro or in vivo and the changes of its tumorigenicity,what should be done firstly is to obtain the transfectant cell lines which express foreign gene .Both recombinant retroviral vector BAG and pXTIL-4(containing LacZ gene and mIL-4 gene, respectively) were used in our study .Packaging cell line Ψ2 was transfected with BAG or pXTIL-4 by calcium phosphate coprecipitation and/or lipofectamine transfectant method.After 2 weeks selecting in medium containing G418,the cell clones— Ψ2BAG and/or Ψ2IL-4 were gained.Fresh retroviral supernatant from the two kinds of cells was used to transfect C6 cell lines,respectively ,and then C6BAG and C6IL-4 cell clone were selected by G418. After being repeatedly freezed and passaged (>10 passages),C6BAG cell could be stained blue by X- gal assay in vitro. β -galactosidase--product of LacZ gene can be expressed stably.The content expressed by Ψ2IL-4 and C6IL-4 in cell culture...
