The carcinogenesis is a multi-step process with multiple factors involved. Because of the genomic instability , under certain environmental condition, tumor cells can escape from immune attack by multiple strategies , such as lack of or reduced expression of MHC antigen and adhesion molecules , and/or secretion of immune suppressive factor(s), so they can grow heterogeneously under the selection force of immune system. Thus, attempts to enhance the immunogenicity of tumor cells and to elicit a potent, long-lasting and specific antitumor immunity have become one of the goals of cancer research.Cytokines have been used in adoptive immunotherapy of tumors for more than 10 years. Facts have demonstrated the antitumor effect of cytokines. However, with very short half-life, cytokines must be administrated with large doses repeatedly to attain therapeutic efficacy, which strongly limits their clinical utilities because of the intolerable systemic toxic side effects . Therefore, a novel therapeutic approach, which includes transducing cytockine genes into tumor cells, inoculating these ' cytokine-producing tumor cells ' to the host , and enabling them to produce and secrete certain amount of cytokines continuously in local environment of tumor, has been developed to reach antitumor...
